TP X2在甲状腺乳头状癌中的表达及其对甲状腺癌KAT细胞增殖能力的影响

摘    要:目的 研究TPX2蛋白(Xklp2靶蛋白)在甲状腺乳头状癌组织中的表达水平及其与临床病理参数的相关性,并探讨其在甲状腺癌KAT细胞增殖过程中的作用.方法 收集我院2013—2016年期间60例新鲜冰冻的甲状腺乳头状癌组织及其配对癌旁组织,采用荧光定量PCR方法检测甲状腺癌组织和癌旁组织中TPX2 mRNA的表达水平,并分析其与临床病理参数的相关性;同时用siRNA干扰甲状腺癌KAT细胞株TPX2的表达,用MTT、流式分析和Western blot方法,分析KAT细胞的增殖能力、凋亡改变及相关凋亡蛋白(cleaved caspase 3、caspase 3)的变化.结果 相对于癌旁组织,甲状腺细胞癌组织中TPX2的表达量在mRNA水平显著高表达(39/60)(P<0.05);在KAT细胞上干扰TPX2表达后,可显著抑制细胞增殖能力,凋亡率增加,同时Cleaved caspase 3、Caspase 3相关凋亡蛋白表达上调(P<0.05).结论 甲状腺细胞癌组织中TPX2表达水平相对癌旁增高,且与甲状腺癌细胞增殖能力相关,可能是甲状腺癌干预的新靶点之一.

Abstract:Objective To investigate the expression of TPX2 in papillary thyroid carcinoma ( PTC) tissues and its correlation with clinical pathological parameters, as well as its effect on KAT cell proliferation. Methods 60 cases of PTC tissues and paired normal tissues were collected in our hospital during 2013-2016. The mRNA level of TPX2 was determined by qRT-PCR. KAT cells were transfected with special siRNA to inhibit TPX2 expression, and then cell proliferation, cell apoptosis and apoptosis-associated protein ( cleaved caspase 3, caspase 3 ) were tested by MTT, flow cytometry and Western blot. Results Compared to the paired normal tissues, TPX2 was highly expressed in tumor tissues at mRNA level ( 39/60) ( P<0.05) . Knockdown of TPX2 effectively reduced KAT cell growth and increased cell apoptosis. Meantime, apoptosis-related proteins cleaved caspase 3 and caspase 3 were up-regulated in TPX2-siRNA-treated group. Conclusion The expression of TPX2 is higher in PTC tissues. TPX2 is highly correlated with cell proliferation, which may be a potential target for treatment of thyroid carcinoma.
