钩藤总生物碱对慢性束缚应激小鼠的抗抑郁作用

摘    要:目的 研究钩藤总生物碱(TAU)对慢性束缚应激(CRS)抑郁模型小鼠的抗抑郁作用.方法 将小鼠束缚在特制的离心管内,每天8 h、连续10 d,制作小鼠抑郁模型,并灌胃给予不同剂量的TAU(80、40、20 mg/kg)和氟西汀(Fluoxetine,FLU 10 mg/kg)治疗.以自主活动实验、强迫游泳实验和糖水偏好实验评价各组小鼠抑郁行为,用ELISA法测定小鼠血清皮质酮(CORT)、肿瘤坏死因子(TNF-α)和NO的含量.结果 TAU能显著增加小鼠体质量,缩短抑郁模型小鼠强迫游泳不动时间及提高小鼠糖水偏好值,改善小鼠抑郁行为;且TAU能显著降低抑郁小鼠血清CORT、TNF-α和NO水平.结论 TAU具有明显的抗抑郁作用,其机制可能是通过调节下丘脑-垂体-肾上腺(HPA)轴功能,减少血清中TNF-α和NO含量.

Abstract:Objective To study the antidepressant effect of total alkaloids from Uncaria ( TAU) in chronic restraint stress ( CRS) mice. Methods Mice were restrained for 8 h daily in a special centrifuge tube for 10 consecutive days to establish the model of depressive mice. The model mice were intragastrically administrated 80, 40 and 20 mg/kg TAU and 10 mg/kg fluoxetine. The behavioral changes were observed by spontaneous activity test, forced swim test and sucrose preference test to evaluate antidepressant effect of TAU. ELISA was used to detect the levels of serum corticosterone ( CORT) , TNF-αand NO. Results TAU significantly increased body weight, shortened immobility period and enhanced sucrose preference in CRS mice. Furthermore, TAU decreased the contents of serum CORT, TNF-αand NO. Conclusion TAU exhibits an antidepressant activity in CRS mice, possibly through regulation of hypothalamus-pituitary-adrenal ( HPA) axis.
