异种骨复合壳聚糖/去甲万古霉素缓释材料的制备及其缓释特性的研究

摘    要:目的 利用静电纺丝技术制备异种骨复合壳聚糖/去甲万古霉素纳米纤维材料,并研究其在体内及体外对去甲万古霉素的缓释性能.方法 以聚乙烯醇(PVA)及壳聚糖(CS)为载体,去甲万古霉素为复合药物,利用静电纺丝技术将纳米纤维膜喷附于异种骨表面及孔隙中;电镜扫描观察纳米纤维膜的大小及形状;建立体肉、体外缓释体系,利用超高效液相色谱(UHPLC)检测不同时间段去甲万古霉素的释放浓度并绘制标准曲线.结果 复合材料电纺纤维膜电镜下质地均匀,直径约50~150nm,在体内、体外缓释实验中均可持续释放有效杀菌浓度抗生素21d以上.结论 异种骨复合壳聚糖/去甲万古霉素纳米纤维材料具有纤维直径小、孔隙率小、比表面积大等特点,可携带更多抗生素并能较长时间持续释放.

Abstract:Objective o prepare xenogeneic bone composite chitosan/norvancomycin nano-fiber material by using electrospinning technique,and study its in vivo and in vitro sustained-release properties to norvancomycin.Methods With polyvinyl alcohol (PVA) and chitosan (CS) as carriers and norvancomycin as compound medicine,nano-fiber membranes was sprayed onto the surface of xenogeneic bone and pores.The size and the shape of nano-fiber mem brane was observed by Scanning Electron Microscope(SEM).In-vivo and in-vitro sustained-release system was set up.The sustained-release density of norvancomycin was detected at different time periods by using ultra-high performance liquid chromatography(UHPLC),and was draw the standard curve.Results The composite material electrospun fiber membranes were homogenous and there was diameter of around 50 to 150nm,and the effective bactericidal concentration of antibiotics can be released continuously for over 21 days in the in-vivo and in-vitro sustained-release tests.Conclusion The xenogeneic bone composite chitosan/norvancomycin nano-fiber material has the characteristics of small diameter,low porosity,high specific surface area,etc.,and can carry more antibiotics and release continuously for long time,which has provided a theoretical basis for the clinical treatment of infectious bone defect.
