抗胰腺癌单克隆抗体的研究

摘    要:用人新鲜胰腺癌组织免疫BALB/c小鼠,取脾细胞与小鼠骨髓瘤细胞Sp2/0-Agl4融合,获得了分泌抗人胰腺癌单克隆抗体的杂交瘤细胞株。本文重点报道以ABC免疫组织化学法检查YPC1单克隆抗体与人正常组织和肿瘤组织的反应性。结果表明YPC1单克隆抗体与16例被检测的胰腺癌组织发生反应。其中9例(56.3%)呈强阳性反应,与23种正常组织中的10种(43.5%)以及19种非胰腺肿瘤中的8种(42.1%)有不同程度的交叉反应,但多数反应较弱。本研究提示以新鲜人体肿瘤组织作为免疫原制备单克隆抗体是简便可行的,YPC1单克隆抗体可能有助于胰腺癌的诊断和治疗。

Abstract:Spleen cells from BALB/c mice, immunized with human pancreatic adenocarcinoma homogenate, were fused with Sp2/0-Agl4 mouse myeloma cells. After screening and cloning, one hybridoma YPC1 which produced monoclonal antibody (McAb) against the original immunizing tumor was selected for further study. When human tissue sections were examined using immunohistochemical technique, YPC1 McAb reacted with all 16 pancreatic adenocarcinoma tissues examined. Nine of these cases (56.3%) showed strong staining. The McAb was also reactive with 10 of 23 (43.5%) normal human tissues and 8 of 19 (42.1%) nonpancreatic tumor tissues. However, except normal tissues including pancreatic duct, and mucosa of the small intestine, colon and bronchus in the lung, and carcinoma tissues from the stomach, colon, bile duct and endometrium, the staining was weak in the remaining cases. These results of the present study suggest that pancreatic carcinoma homo genate is an ideal immunogen for the production of McAb to pancreatic carcinoma and that the YPC1 McAb may be potentially useful for the diagnosis and therapy of pancreatic carcinoma.
