Abstract:Lymphocytes and tumor cells were isolated from pleural effusion of 8 cancer patients by centrifugation on discontinuous density gradients. Cells were incubated at a concentration of 5 x 105 cells/ml in the presence of rIL-2, and tumoricidal activity and phenotypes were assessed at different intervals. The IL-2-activated effusion-associated lymphocytes were found to be propagating well with an increase in cell number up to 60-210 times on the 20th day of incubation. Between Day 10 and Day 15 of incubation, the average cytolytic activities of LAK cells from pleural effusion of 8 cases against K562 and LiBr cells (effector:targct=25:1 ) were 61.6% and 663% respectively, while that of 3 cases against autologous fresh tumor target cells were 46.6%, which were insignificant statistically. Between Day 7 and Day 21 of culture, the cytolytic activities of LAK cells against K562 and LiBr cells kept at a high level, but began to decline by the 21th day. During the whole incubation period of 21 days, the percentage of CD3+ cells kept unvaried while that of CD4+ cells increased and CD8+ cells decreased.
