丹参素对缺氧/缺糖损伤的神经细胞线粒体膜电位和凋亡的影响

摘    要:目的:观察丹参素对神经细胞SH-SY5Y缺氧/缺糖损伤时线粒体膜电位(MMP)和凋亡的影响。方法:将SH-SY5Y细胞分为5组:对照组、模型组、尼莫地平组及丹参素15、30mg/L组,给予相应处理后分别培养2、4、6、12h。四甲基偶氮唑盐(MTT)比色实验检测线粒体活性,流式细胞术检测细胞凋亡百分率和线粒体膜电位,荧光显微镜观察细胞形态学变化和坏死百分率。结果:缺氧/缺糖损伤2h,模型组的线粒体活性和MMP水平较对照组显著降低(P<0.01),并随缺氧/缺糖损伤时间的延长而变化越显著。而细胞凋亡和坏死的百分率均较对照组显著升高(P<0.01),形态学观察也发现染色质凝聚,核碎裂,凋亡小体产生,随着缺氧/缺糖时间的延长,细胞凋亡的发生也越来越多,坏死的细胞也增多。丹参素能显著提高SH-SY5Y细胞线粒体膜电位和活性,降低细胞凋亡及坏死的百分率,其作用随剂量增加而作用增加。结论:丹参素可抑制缺氧/缺糖损伤所致的线粒体膜电位和活性的降低,从而具有稳定线粒体膜电位的作用,抑制神经细胞凋亡的发生。

Abstract:Objective: To investigate the effects of Danshensu on mitochondrion membrane potential(MMP) and apoptosis during injury induced by hypoxia/hypoglycemia in cultured SH-SY5Y cell.Methods: Mitochondrion activity was measured by methyl thiazolyl tetrazolium(MTT)test.The MMP and apoptosis were measured by flow cytometry.Morphological changes and neuronal necrosis were observed with fluorescence microscope.Results: Hypoxia /hypoglycemia cultures for 2 hour significantly declined the MMP and mitochondrion activity(P<0.01),increased the percentage of neuronal apoptosis and necrosis(P<0.01).The phenomena were significance with the extending time of hypoxia /hypoglycemia.The Danshensu could significantly decrease the percentage of neuronal apoptosis and necrosis,increase MMP and mitochondrion activity.With increasing doses of Danshensu,the effects increased linearly with increased dosage.Conclusion: The Danshensu could significantly inhibit the decline in MMP and mitochondrion activity,and had effects of keeping stable of MMP and anti-neuronal apoptosis during injury induced by hypoxia /hypoglycemia.
