| 摘 要： | 为研究我国耶氏肺孢子虫的基因分型,利用巢式PCR方法扩增24例肺孢子虫肺炎(PCP)患者支气管肺泡灌洗液中的耶氏肺孢子虫(Pneumocystis jiroveci)核糖体DNA内转录间隔区(ITS1-5.8S rDNA-ITS2)基因。纯化回收目的片段,将其克隆到PMD18-T载体,并导入JM109细胞系中,每个标本制备3~5个克隆,提取质粒测序。从19例患者的标本中检出ITS1-5.8S rDNA-ITS2基因,其中12份标本的25个克隆的测序结果可用于基因分型。经与Lee等(1998)的分型标准进行比对,本组病例耶氏肺孢子虫的ITS1分为B、E、I、N4型,最常见的为E型;ITS2分为a、b、e、h、i、n6型,最常见的为i型;ITS基因分9型,以Bi多见,有2份标本存在混合感染。

| Abstract: | ITS1-5.8S rDNA-ITS2 regions were amplified from BALF specimens from 24 patients with PCP by use of nested PCR.PCR products were purified,cloned to PMD18-T vector,and introduced to JM109 cell line.ITS of 19 specimens were detected.Of them,25 clones of 12 specimens were available for genotyping.According to the standard of Lee et al.(1998),total of 4 genotypes of ITS1 and 6 genotypes of ITS2 were found.The types of ITS1 were B,E,I,N,and the most prevalent type of ITS1 was E;the types of ITS2 were a,b,e,h,i,n,and the most prevalent type of ITS2 was i.Taken altogether 9 genotypes of ITS were found.The mixed infection was presented from 2 specimens. |
