Abstract:Objectives ①To study the effects of thermal injury and two-hit, induced by endotoxin, on the expression of tumor necrosis factor receptors(TNFR 1 and TNFR2) in intestinal epithelial cells;②To test the hypothesis that rhubarb can inhibit the expression of TNFR1 and TNFR2 in systemic inflammatory response syndrome induced by thermal injury and the two-hit. Methods  Prospective and randomized trial. The rat burn model was used. Endo toxin at a dose of 20 mg/kg was continuously infused 12 hours after burn in rema ining 12 hours. Hybrid ization in situ was applied to evaluate the expression of TNFR1 and TNFR2 in intestinal epithelial cells. The concentration of TNF-α in plasma was alsoobs erved. Results Hybridization in situ revealed that therma l injury and two-hit induced by endotoxin could obviously increase the expression of TNFR1 and TNFR 2 in intestinal epithelial cells. However, the TNFR1 was prevailing. The rhub arb could down-regulate both of the receptors. There was very high concentration of TNF-α in plasma after thermal injury and the two-hit. Rhubarb evidently low ered the concentration of TNF-α in plasma (P<0.01). Conclusion  Rhubarb could down-regulate the expression of TNFR1 and TNFR2 in intestinal epithelial cells after thermal injury and the two-hit. It also decreased the concentration of TNF-α in plasma.
