CGRP对人骨髓间充质干细胞增殖影响的研究

摘    要:目的探讨外源性重组人降钙素基因相关肽(hCGRP)对人源性骨髓间充质干细胞(human mesenchymal stem cells,hMSCs)的增殖的影响,进而探索CGRP促进成骨的作用机制。方法采用梯度离心法和选择性培养基分离、纯化骨髓间充质干细胞,培养体系中添加不同浓度CGRP。于传代第三代进行细胞鉴定,光镜下观察细胞形态、MTT法检测细胞增殖能力、流式细胞仪测定细胞周期,以观察CGRP对MSCs增殖能力的影响。结果CGRP对MSCs增殖形态无明显影响,MTT示CGRP组细胞增殖速率快于对照组,流式细胞仪检测示CGRP组的MSCs的G2/G1期比例明显升高。结论CGRP对MSCs的增殖有直接促进作用,从而有利于骨形成。

Effect of Calcitonin Gene Relate Peptide on Proliferation of Human Mesenchymal Stem Cells

Abstract:Objective To explore the effect and the mechanism of recombinant human calcitonin gene relate peptide (hCGRP) on proliferation of human mesenchymal stem cells (hMSCs). Methods The hMSCs were isolated from healthy volunteers' bone marrows with gradient centrifugal methods, and treated with different concentration of CGRP. Cells of the third generation were identified with cells' surface marker. Cells' morphology was observed with optical microscope, proliferation was detected with MTT exam, and cell cycle was determined with FCCS. These results were used to evaluate the effect of CGRP on the proliferation of hMSCs. Results CGRP can enhance the proliferation of hMSCs significantly. The ratio of G2 to G1 was raised in the CGRP group. Conclusion CGRP can increase bone formation by promoting the proliferation of MSCs directly.
