盐酸戊乙奎醚对哮喘鼠气道平滑肌细胞增殖作用的研究

摘    要:目的探讨盐酸戊乙奎醚对哮喘鼠气道平滑肌细胞(ASMC)增殖的影响。方法BALB/c小鼠随机分为对照组(A)、哮喘组(B)和盐酸戊乙奎醚干预组(C)。进行支气管肺泡灌洗液(BALF)中细胞分类计数;测定转移生长因子-β1(TGF-β1)的浓度及增殖细胞核抗原(PCNA)的表达;观察肺组织病理形态学变化;图像分析测定支气管基底膜周径(Pbm)、气道壁总面积(WAt)、平滑肌面积(WArn)。结果C组的TGF-β1、PCNA表达及WAt/Pbm、WAm/Pbm比值明显低于B组(P〈0.05),而高于A组(P〈0.05)。结论早期行盐酸戊乙奎醚干预可通过减少TGF-β1在肺组织的表达减轻ASMC增殖。

Abstract:Objective To explore the effects of Penehyclidine Hydrochloride on the proliferation of airway smooth muscle cells(ASMC) of chronic asthma mice. Methods BABL/e mice were divided into 3 groups randomly: group A (control group), group B ( asthma group), group C ( Penehyclidine Hydrochloride - intervention group). Total and differential cell counts in bronchoalveolar lavage fluid (BALF) were observed. To detect the concentration of active TGF -β1 and proliferating cell nuclear antigen (PCNA) and the pathomorphological changes in the lung were analyzed. The perimeter of basement membrane(Pbm) ,the total bronchial wall area (WAt) , the muscle wall area (WAm) were measured by using image analysis system. Results TGF-β1, PCNA level and WAm/Pbm, WArn/Pbm and WAi/Pbm of group C decreased greatly in comparison with group B ( P 〈 0. 05 ), but increased obviously in comparison with group A ( P 〈 0.05 ). Conclusion Early intervention with Penehyclidine Hydroehloride might partially inhibits its process via higher expression of TGF -β1 in chronic asthma and decrease the proliferation of ASMC.
