摘    要:目的研究5-烯丙基-7-二氟亚甲基白杨素(ADFMChR)对体外培养人肺癌(A549)细胞、中国仓鼠肺上皮(CHL)细胞增殖活性的影响。方法体外培养A549细胞和CHL细胞,MTT比色法测定细胞增殖活性。结果ADFMChR在终浓度为1.0、2.0、4.0、8.0、16.0μmol/L时,处理A549细48 h,细胞增殖抑制率分别为24.39%、47.66%、62.75%、73.35%、85.94%,其抑制率高于先导化合物(白杨素),而与顺铂(DDP)相当,且呈浓度依赖性。终浓度为1.0、2.0、4.0、8.0、16.0μmol/L的ADFMChR处理中国仓鼠肺上皮CHL细胞48 h,细胞增殖抑制率为3.95%、7.44%、13.89%、21.60%、27.43%,其对CHL细胞的毒性作用与白杨素相当,而低于DDP。结论ADFMChR对肺癌A549细胞增殖抑制作用强,而对中国仓鼠肺上皮CHL细胞的抑制作用较弱,其抑制A549细胞增殖作用具有选择性。

Abstract:Objective To investigate the effects of 5 - allyl - 7 - gen - difluoromethyllenechrysin ( ADFMChB ) the proliferative viability of cultured human lung cancer cell line (A549) and Chinese hamster lung epithelial cell( CHL ) in vitro. Methods A549 cell line and CHL cell line were cultured in vitro. The viability inhibition of two cell line were examined by MTT assay. Results The MTT assay showed that the proliferation inhibitory rates of A549 cell line were 24.39% ,47.66 % ,62.75 %, 73.35 %, 85.94% respectively after treatment with 1. 0, 2. 0, 4. 0, 8.0,16. 0 μmol/L of ADFMChR for 48h. ADFMChR significantly suppressed proliferation of A549 cell line in a concentration dependent manner. The efficacy of inhibition with ADFMChR was stronger than that of Chrysin while it was similar to DDP. For CHL cell line, the proliferation inhibitory rates were 3.95% ,7.44%, 13.89% ,21.60% ,27.43% respectively after CHL cell was treated with 1.0,2.0,4.0,8.0,16.0 μmol/L of ADFMChR for 48 h. The eytotoxieity of ADFMChR to CHL cells was lower than that of DDP while it was similar to Chrysin. Conclusions ADFMChR had stronger cell proliferation inhibition effect on A549 cell line than to CHL cell line and it possessed the relative selectivity for cell proliferation inhibition on A549 cell.
