中药复方不同提取方法药效成分动态变化研究

摘    要:目的:本实验以葛根芩连汤不同提取方法的提取液作为研究对象,采用高效液相色谱方法,在不同色谱条件下对中药复方药效成分进行测定,通过指纹图谱对比及指标性成分测定探讨其动态变化。方法:采用HPLC方法,选用ODS色谱柱(200mm×4.6mm,5μm),流动相为甲醇-0.02mol.L-1磷酸二氢钾0.2%磷酸水溶液(pH=3.0)梯度洗脱,流速为1.0mL.min-1,检测波长250nm,建立葛根芩连汤的HPLC指纹图谱;分别测定方中主要指标成分葛根素、盐酸小檗碱、黄芩苷、甘草酸的含量,并通过SPSS16.0统计软件计算含量的差异性。结果:葛根芩连汤不同提取液中化学成分在质上未有明显变化,但在含量上的变化比较显著。结论:不同提取方法影响复方中药效成分的含量及组成变化。

Abstract:Objective: This experiment,Gegenqinlian Decoction extract of different extraction methods as an object of study,using high performance liquid chromatography method,under different chromatographic conditions were components of Chinese medicine compound efficacy was determined by fingerprint comparison and indicators for measurement of ingredients to explore its dynamics.Methods: HPLC method,use ODS column(200mm×4.6mm,5μm),mobile phase was methanol-0.02mol / l potassium dihydrogen phosphate 0.2% phosphoric acid solution(pH=3.0) gradient elution,flow rate 1.0ml/min The detection wavelength was 250nm,the establishment of Gegenqinlian Decoction of the HPLC fingerprint;Fangzhong key indicators were measured puerarin,berberine,baicalin,glycyrrhizin content,and through SPSS16.0 statistical software to calculate differences in content.Results: Gegenqinlian Decoction extract of a different chemical composition in terms of quality did not significantly change,but the content more prominent on the change.Conclusions: Different extraction methods affect the efficacy of compound in the composition changes in the content and composition.
