摘    要:用流式细胞术(FCM)测定了45例不同病期急性白血病患儿和13例非白血病患儿骨髓细胞原位DNA、RNA和蛋白质含量,对急性淋巴细胞白血病(ALL)治疗前、急性非淋巴细胞白血病(ANLL)治疗前、急性淋巴细胞白血病缓解期(CR)和非白血病患儿共四组病人的上述各项指标进行了分析。结果显示:1.急性白血病患儿的DNA非整倍体检出率为41.2%,其中ALL为33.3%,ANLL为60%。ALL和ANLL的S期细胞百分比(S%)显著低于非白血病患儿,而CR组的S%与非白血病的S%无显著性差异。2.ANLL组的RNA含量显著高于CR组、非白血病组及ALL组,而后三组间无显著性差异。3.四组之间的蛋白质含量无显著性差异。4.在对9例缓解期病人进行动态观察过程中,有5例检出DNA非整倍体,其中4例在检出DNA非整倍体后33~74天内复发。

DETERMINATION OF DNA、RNA AND PROTEIN CONTENT OF BONE MARROW CELLS IN CHILDHOOD ACUTE LEUKEMIA BY FLOW CYTOMETRY(FCM)
Abstract:Abstract:DNA, RNA and protein content of bone marrow mononuclear cells from 45 cases of acute leukemia and 13 cases of non-leukemic chidren were determined simultaneously by flowcytometry using PI+FITC and PY+MG stainings. The overall incidence of DNA aneuploidyin acute leukemia was 41. 2%. No diffrence of DNA aneuploidy in acute leukemia was 41.2%. No diffrence in the frequence of DNA aneuploidy was found between ALL (33. 3% )andANLL (60% ). The percentage of S phase (S%)of bone marrow cells from ALL and ANLLwas significantly lower than that from CR and non-leukemic children. The content of RNAin ANLL patients' bone marrow cells was significantly higher than that in ALL CR and nonleukemic children. There were no significant differences in LPCF and protein content from 4groaps. In the consecutive observation of 11 cases of CR patients, 5 cases were found to haveDNA aneuploid cells,4 of them relapsed in 33 to 74 days after DNA aneuploid cells beingfornd.
