兔下颌骨髁状突软骨细胞体外培养及生物学特性的研究

摘    要:为探讨兔下颌骨髁状突软骨细胞的体外培养方法及其生物学特性,用胰酶及胶原酶联合消化法获取软骨细胞,并在DMEM培养基中进行原代及传代培养;通过形态学观察及免疫组织化学染色对软骨细胞的贴壁率、生长曲线及表型等细胞生物学特性进行了研究。结果:所获软骨细胞主呈多角形,第3代软骨细胞12h贴壁率达95%,软骨细胞倍增时间约55.1h,6代以内软骨细胞的Ⅱ型胶原蛋白稳定表达。结果表明:本方法获软骨细胞具有稳定

Abstract:To investigate the biological behaviors of rabbit condylar chondrocytes, the primary chondrocytes were harvested from newborn rabbits by dissection and sequential digestion with 0 25% trypsin and 0 l % collagenase IA The isolated chondrocytes were cultured in DMEM medium, their morphology, growth and phenotype were observed by phase-contrast microscope and immunohistochemical method Results: The chondrocytes displayed polygonal dominated and the positive result of collagen 11 within six passage Conclusion: The chondrocytes can kept steady proliferation and their phenotype in this culture condition
