人胰腺腺癌中抗癌基因P53的突变

用ＰＣＲ－ＳＳＣＰ分析及ＤＮＡ直接测序法检测了１６例胰腺腺癌的Ｐ５３基因５－８外显子，结果发现３７．５％有Ｐ５３基因点突变，突变分布于外显子５－８，无特殊突变倾向点。６例有Ｐ５３突变者，５例为１个等位基因突变，另１个等位基因丢失；１例同时含有突变型和隆型等位基因。

P53 mutations in pancreatic adenocarcinoma

Using a combination of the single-strand conformation polymorphism and direct sequencing of PCR-amplified fragments, we analyzed 16 pancreatic adenocarcinoma samples for the presence of mutations in exons 5, 6, 7 and 8 of the P53 gene.Thirty-eight percent (6/1 6) of the pancrcatic adenocarcinomas tested presented a mutated P53 allele.Mutations were located in exons 5 to 8, no specific site seemed to be preferentially affected. In five tumors the mutation of one allele was concomitant with the loss of the wild-type counterpart. Another sample presented both the mutated allele and the wild-type allele.The transitions were four G to A and one A to G ; the transversions were one A to C.Only one mutation was transition occurring at CpG dinucliotide site.Our data suggest that P53 mutation may have a role in the pathogenesis of pancreatic cancers.
