Vero细胞检测出血性大肠杆菌O157:H7毒素的方法研究

目的建立用Vero细胞检测大肠埃希菌毒素的方法。方法用两种不同的培养基培养10株大肠杆菌和1株痢疾志贺杆菌,将其培养物上清和菌体裂解液加入Vero细胞中,观察对Vero细胞的毒性作用。结果两种培养基培养大肠杆菌产生的上清对Vero细胞的毒性作用差异不大,大肠杆菌培养上清的毒性作用大于菌体,而痢疾志贺菌菌体的毒性作用大于上清。结论Vero细胞毒性检测方法可以用于检测出血性大肠杆菌细胞毒素。

A method using Vero cells to detect EHEC(O157:H7) toxins

Objective To develop a new method using Vero cells to detect E.coli toxins.Method 10 E.coli strains and one Shigella dysenteriae strain were cultured using two different culture mediums and the supernatant and bacteria lysis were put into Vero cells.The toxic response of the Vero cells was observed.Result No difference was found between the two type of supernatant using two different culture mediums in the toxic response to Vero cells.Toxicity of supernatant of E.coli was more great than bacteria lysis.Toxicity of bacteria lysis of Shigella dysenteriae was more great than the supernatant.Conclusion The method using Vero cells can be used to detect EHEC toxins.
