摘    要:目的:研究幽门螺杆菌(Hp)中国菌株转位因子IS605的序列差异。方法:采用PCR扩增、克隆、连接、测序的方法获得不同地区菌株IS605的序列并进行聚类分析比较。结果:中国菌株IS605的核苷酸序列高度保守,不同地区同源性达93%,相同地区同源性达95%,G C含量为35%,与Hp的全基因相似。同源聚类分析表明相同地区IS605的序列相近。结论:IS605是幽门螺杆菌基因组中相当保守的成分,其移位的机制有待于研究。

Abstract:Objective To analyze the conservation and diversity of IS605 of Helicobacter pylori in China. Methods PCR amplification and the PCR products were cloned into pGEM T vector and then sequenced respectively. The sequences were compared by Vector NTI Suite 6.0. Result The DNA sequence of IS605 is highly conserved. Their G+C contents were 35% similar with other genes in H.pylori . The DNA sequences of IS605 positive strains from the same region are highly homogeneous. Conclusion IS605 may be one ancient component of the Helicobacter pylori gene pool. We need advanced study to access the mechanisms of their transposition. [
