生长激素对脐血来源树突细胞成熟状态的影响

摘    要:目的 检测生长激素(GH)对脐血来源树突细胞(DC)的免疫表型和功能状态的影响.方法 从新鲜脐带血中分离出单核细胞,应用白细胞介素4(IL-4)和重组人粒-巨细胞集落刺激因子(rhGM-CSF)诱导刺激单核细胞,以不同浓度的GH作为修饰因子修饰DC诱导过程;观察DC形态、数目、生长状态,流式细胞仪检测DC的免疫表型,酶联免疫吸附试验测定DC上清液中白细胞介素12(IL-12),自体混合淋巴细胞反应测定其功能状态.结果 脐血贴壁的单核细胞在细胞因子和GH诱导下可以分化为成熟DE,不同浓度(5、25、100 ng/ml)GH可显著上调DC CD83(31%±4%、34%±4%、34%±5%)、CD80(26%±4%、29%±6%、30%±6%)和HLA-DR(45%±8%、49%±6%、50%±5%)分子表达的阳性宰(P<0.05),明显提高DC分泌IL-12的能力[分别由第5天的(91±12)、(159±20)、(166±19)pg/ml上升到第9天的(686±58)、(796±61)、(810±67)pg/ml](P<0.05),DC对自体淋巴细胞的激活功能明显增强(P<0.05).结论 GH可明显促进DC的诱导过程,修饰加速DC的成熟并提高细胞表面共刺激分子表达,增强DCIL-12分泌和激发自身淋巴细胞增殖能力;GH的修饰作用与浓度有一定的相关性.

Abstract:Objective To detect the effect of functional status and immunophenotype of dendritic cell modified witll growth hormone(GH)from cord blood.Methods Monocyte wa8 isolated from fresh cord blood.DCs were induced with IL-4 & GM-CSF and medified with different concentrations of GH.The morphology,count and growth status of DC were observed appearance.And the immunophenotype of DC was detected with flow cytometry.IL-12 in supernatant wa8 measured with ELISA.The inununo-activity was determined with autologous mixed lymphocyte reaction.Results Mature DC were induced from adherent lymphomonoeyte under the action of IL-4.GM-CSF and GH of different densities(5,25,100 ng/ml).GH increaes the positive expression of CD83(31%±4%,34%±4%,34%±5%),CD80(26%±4%,29%±6%,30%±6%)and HIA-DR(45%±8%,49%±6%,50%±5%)(P<0.05),enhanced theability of secreting IL-12(P<0.05)and boosted significantly the immuno-activation of DC to autologous lymphocyte(P<0.05).Conclusion GH could promotethe differentiation,maturationand expression of co-stimulatory molecules,increase the IL-12 production in DC and enhance the ability of DC to stimulate the proliferation of lymphocyte within a certain dose range.
