龙血素A对瘢痕疙瘩成纤维细胞生长作用研究

摘    要:目的:研究不同浓度龙血素A对瘢痕疙瘩成纤维细胞增殖、凋亡、迁移能力的影响以及对Col-I、Col-III、α-SMA、Bax、Caspase-3蛋白表达水平的影响。方法:1)不同浓度龙血素A作用于瘢痕疙瘩成纤维细胞,用CellTiter-Blue活力检测试验检测龙血素A对瘢痕疙瘩成纤维细胞增殖的影响。2)流式细胞术检测龙血素A对瘢痕疙瘩成纤维细胞凋亡的影响。3)划痕实验检测龙血素A对瘢痕疙瘩成纤维细胞迁移的影响。4)Western-Blot测定龙血素A对瘢痕疙瘩成纤维细胞Col-I、Col-III、α-SMA、Bax、Caspase-3蛋白水平的影响。结果:龙血素A可抑制瘢痕疙瘩成纤维细胞增殖、迁移并诱导其凋亡,同时减少COL-I、COL-III、α-SMA蛋白表达,并促进Bax、Caspase-3蛋白表达,并且均呈现剂量依赖性。结论:龙血素A可以调控瘢痕疙瘩成纤维细胞增殖与凋亡的平衡、减少细胞外基质沉积,从而可能对KD发挥一定的治疗作用。

Abstract:Objective:To study the effects of different concentrations of Loureirin A on the proliferation,apoptosis and migration of keloid fibroblasts and the protein expression levels of Col-I、Col-III、α-SMA、Bax、Caspase-3.Methods:1)Different concentrations of L-Auxin A act on keloid fibroblasts.CellTiter-Blue vitality test was used to test the effects of L-Auxin A on keloid fibroblast proliferation.2)Flow cytometry was used to detect the effects of L-Auxin A on keloid fibroblasts apoptosis.3)Scratch test was used to detect the effects of borneol A on keloid fibroblast migration.4)Proteins expression levels of Col-I、Col-III、α-SMA、Bax、Caspase-3 were detected by Western-Blot.Results:Loureirin A can inhibit the proliferation and migration of keloid fibroblasts,and induce their apoptosis,reduce the protein expression of Col-I、Col-III、α-SMA and promote the protein expression of Bax and caspase-3,and there is a dosage dependent relationship.Conclusion:Loureirin A can regulate the balance of keloid fibroblast proliferation and apoptosis,reduce the deposition of extracellular matrix,so it may play a certain therapeutic effects on keloid.
