舒血宁联合磷酸肌酸钠治疗对心肌梗死大鼠的心肌保护作用及对血管再生的影响

摘    要:目的:探究舒血宁联合磷酸肌酸钠对心肌梗死大鼠的心肌保护作用及对血管再生的影响。方法:将60只建模成功的心肌梗死大鼠随机分为模型组、磷酸肌酸钠及联合组,每组20只,另选20只SD大鼠仅于左冠状动脉前降支穿线而不结扎作为假手术组。磷酸肌酸钠及联合组分别腹腔注射磷酸肌酸钠[0.2 g/(kg·d)]、磷酸肌酸钠[0.2 g/(kg·d)]+舒血宁[10 mL/(kg·d)],模型组及假手术组则注射等体积生理盐水,各组均连续干预4周。检测并比较各组大鼠心功能指标;观察各组大鼠心肌组织病理学形态变化;比较各组大鼠心肌组织微血管密度及血管内皮生长因子(VEGF)表达情况。结果:模型组大鼠左心室舒张末期内径(LVEDd)、左心室收缩末期内径(LVESd)较假手术组升高,而磷酸肌酸钠组及联合组较模型组降低,且联合组显著低于磷酸肌酸钠组,差异有统计学意义(P<0.05);模型组大鼠左心室短轴收缩率(FS)及左心室射血分数(LVEF),心肌组织CD31、VEGF蛋白A值及MVD较假手术组降低,而磷酸肌酸钠组及联合组较模型组升高,且联合组显著高于磷酸肌酸钠组,差异有统计学意义(P<0.05)。结论:舒血宁联合磷酸肌酸钠有效促进心肌组织缺血区域血管新生,改善心肌梗死大鼠心肌病理损伤,恢复其心功能,疗效显著优于单用磷酸肌酸钠。

Abstract:To explore the myocardial protection of Shuxuening combined with sodium phosphate in the treatment of myocardial infarction in rats and the effect on angiogenesis.Methods:A total of 60 cases successfully modeling myocardial infarction rats were divided randomly into a model group, a sodium phosphate group and a combined group, with 20 cases in each group, and another 20 SD rats were only in the left anterior descending coronary artery threading without ligation and acted as sham-operation group.The sodium phosphate group and the combined group treated with intraperitoneal injection of sodium phosphate 0.2 g/(kg·d), sodium phosphate 0.2 g/(kg·d)+ Shuxuening 10 mL/(kg·d)respectively.The model group and the sham-operation group were treated with intraperitoneal injection isovolumetric saline.Each group was treated for 4 weeks continuously.The rat cardiac function indexes in each group were detected and compared.The myocardial tissue pathology morphological changes were observed in each group.The myocardial tissue microvascular density and VEGF protein expression in each group were detected and compared.Results:The LVEDd, LVESd of rats in the model group were higher than those in the sham-operation group, while the sodium phosphate group and the combined group were lower than the model group, and the combined group was lower than the sodium phosphate group(P<0.05); the FS, LVEF, IOD values of myocardial tissue CD31, VEGF protein and MVD in the model group were lower than those in the sham-operation group, while the sodium phosphate group and the combined group were higher than the model group, and the combined group was higher than the sodium phosphate group(P<0.05).Conclusion:Shuxuening combined with sodium phosphate can promote the myocardial tissue ischemia region of angiogenesis effectively, improve the myocardial pathological damage of myocardial infarction rats, and restore the heart function.The effect is better than sodium phosphate alone significantly.
