Abstract:Objective To study the protective effect of grape procyanidins(GPC) on the proliferative activity of mouse hepatocytes damaged by ethanol. Methods The mice were randomly divided into five groups,Those in low-,moderate-and high-dose GPC groups were intragastrically given 50,100 and 200 mg/kg of GPC,and those in ethanol group,4 g/kg of ethanol were given,respectively,for four weeks.All the mice were then sacrificed.The liver cell multiplication activity was determined by MTT assay,the pathology of liver tissue section stained by HE was observed light-microscopically;the expressions of Bcl-2 and Bax protein in liver cells were measured by immunohistochemistry method;the serum alanine aminotransferase(ALT) was detected by automatic biochemical analyzer,and the serum TNF-α was detected by ELISA method. Results The ALT levels of high-dose GPC group were lower than that of the ethanol group(F=14.05;q=4.29,5.53;P<0.05),and the cell proliferation activity was higher(F=68.68;q=5.71,8.57;P<0.05).Compared with the ethanol group,the ratio of Bcl-2/Bax was significantly higher in the high-dose GPC group(F=388.03;q=4.90;P<0.05),the TNF-α levels of the high-and moderate-dose GPC groups were lower(F=10.09;q=3.01,5.68;P<0.05).GPC improved the liver injury in different degrees. Conclusion GPC can prevent alcohol-induced injury of liver cell multiplication activity by regulating the ratio of Bcl-2/Bax and lowering serum TNF-α concentration.
