Bcl-xL、Bcl-Xs/L以及Bcl-6在骨肉瘤组织中的表达及临床意义

目的:检测凋亡相关蛋白Bcl-xL、Bcl-Xs/L以及Bcl-6在骨肉瘤中的表达,并探讨其与骨肉瘤预后的关系。方法:应用免疫组化方法检测Bcl-xL、Bcl-Xs/L以及Bcl-6在41例骨肉瘤组织中的表达,并分析其与无瘤生存率及生存时间的关系。结果:Bcl-xL、Bcl-Xs/L在骨肉瘤组织中表达的阳性率分别为53.7%和75.6%,而Bcl-6为阴性表达,是否行术前化疗不影响Bcl-xL的表达(P>0.05),Bcl-xL表达阳性者无瘤生存率较阴性者低(P<0.01),而Bcl-Xs/L表达与复发转移及无瘤生存率无相关关系。结论:Bcl-xL、Bcl-Xs/L在骨肉瘤中有不同程度的表达,Bcl-xL的表达与预后相关,Bcl-xL阳性表达者复发、转移率高,无瘤生存率低,这与Bcl-xL抑制骨肉瘤细胞凋亡导致化疗耐药的特性相关。

Bcl-xL, Bcl-Xs/L, Bcl-6 expressions in osteosarcoma and clinical significance

Objective:To investigate the expressions of Bcl-x L ,Bcl-Xs /L and Bcl-6and their re-lationship with clinical outcome.Methods:Tumor biopsies from41patients with osteosarcoma were analyzed with immunohistochemistry,using antibodies specific for Bcl-x L ,Bcl-Xs /L and Bcl-6.Clinical outcome,disease-free survival were compared among the three groups.Results:The expression rate of Bcl-x L and Bcl-Xs /L was53.7%and75.6%respectively.The expression of Bcl-x L correlated with poor disease-free survival,but the expression of Bcl-Xs /L was not found to correlate with the clinical out-come.Bcl-6was not expressed in all cases.Conclusion:The expression of Bcl-x L correlates with poor prognosis.Patients with tumors of positive Bcl-x L expression appear to have strong likelihood of relapse and metastasis and low disease-free survival rate.This may be attributed to the mechanisms of apopto-sis inhibition and chemotherapy resistance of Bcl-x L .
