咪唑立宾对急性Thyl肾炎大鼠细胞周期调控蛋白CyclinD1的抑制作用

摘    要:目的探讨咪唑立宾对急性Thyl肾炎大鼠肾小球系膜细胞增殖的抑制作用及其对细胞周期调控蛋白CyclinD1的影响。方法54只雄性Wistar大鼠随机分为对照组、Thyl肾炎模型组和咪唑立宾组各18只。模型组与咪唑立宾组尾静脉注射OX-7细胞上清浓缩液制备急性Thyl肾炎模型,对照组注射等量生理盐水。注射后6h灌胃给药,对照组与模型组给予蒸馏水,咪唑立宾组给予咪唑立宾20mg/kg,连续给药7d后,检测3组24h尿蛋白定量,采用PAS染色评估肾脏组织病理改变,应用RT—PCR法检测CyclinD1基因表达情况,应用Westernblot法检测CyclinD1蛋白表达情况。结果模型组24h尿蛋白定量、肾小球内细胞数、CyclinD1mRNA及CyclinD1蛋白表达水平均明显高于对照组和咪唑立宾组(P〈0.05)。结论咪唑立宾通过调控细胞周期调节蛋白CyclinD1抑制急性Thyl肾炎大鼠系膜细胞增殖,减少尿蛋白。

Abstract:Objective To explore the inhibitory effect of mizoribine on the proliferation of mesangial cell and the influence on the cell cycle regulatory protein Cyclin D1 in rats with acute Thyl nephritis. Methods Fift-four rats were randomly divided into control group, nephritis group and mizoribine group, with 18 rats in each group. Both nephritis group and mizorihine group received caudal vein injection of OX-7 cell supernatant concentrate to establish Thyl nephritis rat models, and control group was injected the same volume of saline. Control group and nephritis group were treated with distilled water after 6 hours, while mizoribine group was treated with 20 mg/kg of mizoribine for totally 7 days. Seven days after treatment, all three groups were detected 24-hour urinary protein excretion, assessed the pathological changes by PAS, detected the expression of Cyclin D1 mRNA by RT-PCR, and detected the expression of Cyclin D1 protein by Western blot. Results Seven days later, the 24 hour urinary protein excretion, the number of glomerular cells, and the expression levels of Cyclin D1 mRNA and Cyclin D1 protein were significantly higher in nephritis group than those in control group and mizoribine group (P〈0. 05). Conclusion Mizorihine can inhibit mesangial proliferation and reduce urine protein in acute Thyl nephritis rats by up-regulating the expression of Cyclin D1 protein.
