摘    要:目的观察益母草水苏碱(stachydrine,Sta)对去甲肾上腺素(norepinephrine,NE)诱导的新生大鼠心肌细胞胚胎基因再表达的抑制作用。方法采用II型胶原酶分离新生大鼠心肌细胞,随机分为空白对照组、NE组、卡维地洛组(Car组)及益母草水苏碱组(sta组)各组分别予相应的干预措施。采用荧光标记F-actin染色法测定心肌细胞表面积,Protein/DNA比值代表细胞蛋白合成,并以荧光定量PCR检测胚胎基因α—MHC、β-MHCmRNA表达情况。结果与空白对照组比较,NE组心肌细胞表面积、Protein/DNA比值以及3-MHCmRNA表达差异有统计学意义(P〈0.05)。与NE组比较,sta组及卡维地洛组心肌细胞表面积、Protein/DNA及β—MHCmRNA表达差异有统计学意义(P〈0.05)。结论Sta能够显著抑制NE诱导的心肌细胞肥大。

Inhibitory effects of Stachydrine of Leonurus on cardiaomyocyte hypertrophy induced by norepi- nephrine

Abstract:Objective To observe the effects of Stachydrine of Leonurus on cardiac myocyte hypertrophy induced by norepinephrine. Methods Neonatal rat ventrieular cardiomycytes were isolated from 1-2 day-old-rat by collagenase type II enzyme. Cardiac myocytes were then treated with norepinephrine ( 1 μmol/L) for 72h in the presence or absence of earveditol ( as a positive control, 1μmol/L), or stachydrine ( 10 μmol/L), while controls left in medium without treatment. For cell surface area measurement, cardiac myocytes were labeled with Alexa Fluor 488 conjugated phalloidin and observed by a fluorescent microscopy. Protein synthesis was evaluated by the ratio of Protein/DNA. Where Protein concentration was determined by BCA protein assay; DNA concentration was determined with Hoescht 33258 by calf thymus DNA as a standard. The mRNA expression of two isoforms of myosin heavy chain (MHC), α-MHC and β-MHC were measured by Real-time PCR. Results NE significantly increased the cell area, protein content, Protein/DNA ratio, β-MHC expression and β-MHC ratio in cultured cardiomyocyte ( P 〈0. 05 ). While these changes were significantly abrogated by stachydrine ( P 〈 0.05 ). Conclusion Staehyddne attenuate NE-induced cardiomyocytes hypertrophy.
