摘    要:目的:探讨微小染色体维持蛋白2(MCM2)和增殖细胞核抗原(PCNA)在宫颈癌组织中的表达及生物学意义。方法:采用免疫组化ABC法检测MCM2和PCNA在不同病变宫颈组织芯片中的表达。结果:MCM2、PCNA在宫颈癌组织中的表达高于正常宫颈组织,差异有显著性(P〈0.05)。PCNA表达在中分化癌组高于高分化癌组,差异有显著性(P〈0.05)。MCM2和PCNA的标记指数(LX)随宫颈病变进展而增加。其中MCM2 LI和PCNA LI分别在宫颈癌组与宫颈炎组的比较差异有显著性(P〈0.001和P〈0.01)。比较宫颈癌组和CIN组中MCM2 LI和PCNA LI差异也具有显著性(P〈0.05和P〈0.01)。结论:MCM2和PCNA在宫颈癌变过程中起促进作用。与PCNA相比,MCM2是更敏感、可靠地反映宫颈上皮细胞增殖的指标,对判断宫颈病变的性质及程度有参考价值。

Abstract:Objective To explore the expressions of MCM2 and PCNA in cervical cancer and their biological significance.Methods The expressions of MCM2 and PCNA in the cervical tissue microarray were detected by immunohistochemical ABC methods.Results The expression levels of MCM2 and PCNA in cervical cancer were significantly higher than those in normal cervical tissues(P < 0.05).PCNA expression was higher in moderate-differentiated than in well-differentiated cancer(P < 0.05).The average MCM2 and PCNA labeling indices(LIs) elevated with the progression of the cervical lesions and differed significantly between cervical cancer and cervicitis(P < 0.001 and P < 0.01).There were significant differences in MCM2 and PCNA LIs between cervical cancer and cervical intraepithelial neoplasia(P < 0.05 and P < 0.01).Conclusions Both MCM2 and PCNA play a promoting role in the carcinogenesis of cervical cancer.MCM2,as compared with PCNA,is a more sensitive,reliable indicator of epithelial proliferation of the cervix,and may be useful in judging the nature and degree of cervical lesions.
