黄萱益肝散抗肝损伤及肝细胞凋亡的实验研究

目的:研究黄萱益肝散抗肝损伤及肝细胞凋亡的作用。方法:以武汉麻鸭(感染鸭乙型肝炎病毒)为鸭乙型肝炎动物模型,30只感染鸭乙型肝炎病毒的武汉麻鸭随机分为模型组、拉米夫定组和黄萱益肝散组(又分大、中、小剂量组);分别在给药第14 d及停药后第3 d采血,检测谷丙转氨酶/谷草转氨酶(ALT/AST);在停药后第3 d处死各组麻鸭后,取小块肝脏组织,光镜观察肝脏病理情况;TUNEL法检测肝细胞凋亡情况。结果:黄萱益肝散组在给药第14 d、停药后第3 d的ALT/AST较模型组有所下降(P<0.05,P<0.01),其肝细胞变性、坏死以及炎症等病理变化较模型组有好转,肝细胞凋亡指数较模型组也有所下降(P<0.05,P<0.01)。结论:黄萱益肝散有保护肝细胞和抑制肝细胞凋亡的作用。

Experimental Study of Huang Xuan Yi Gan San on Liver Injury and Hepatic Cell Apoptosis

Objective:To study the effect of Huang Xuan Yi Gan San on liver injury and hepatic cell apoptosis.Me-thods: Wuhan brown spot ducklings infected with D-HBV were used as the hepatitis B animal model,thirty ducks were randomly divided into model group,lamivudine group and Huang Xuan Yi Gan San group.Blood samples were taken from each duck on the first day before treatment,on the day 14 after treatment and on the day 3 after recession of treatment,ALT and AST were detected.All the ducks were killed on the day 3 after recession of treatment and taken the fresh liver tissues,the pathological structures were observed,the apoptosis rate was detected with TUNEL.Results: The levels of ALT and AST in the Huang Xuan Yi Gan San group were obviously lower than those of the model group after medication(P<0.05,P<0.01).The hepatic pathological structures in the Huang Xuan Yi Gan San group turned good and the apoptosis rate was significant difference compared with the model group(P<0.05,P<0.01).Conclusion: Huang Xuan Yi Gan San can protect the hepatic cell and inhibit hepatic apoptosis.
