不同转移潜能结肠癌细胞比较性实验研究

目的:评估影响结肠癌进展的主要因素,为临床有效防治提供实验依据。方法:对两种具有不同转移潜能的人结肠癌细胞株LS174T及HCT8,作生长能力、侵袭力、癌细胞表面HLA及共刺激分子表达、诱导血管内皮细胞迁移能力、血管内皮生长因子表达等指标的检测。结果:两种癌细胞株主要在诱导血管内皮迁移能力上存在不同,LS174T及HCT8诱导的内皮细胞迁移数目分别为154.6±37.9个及113.0±72.7个,差异有统计学意义(t=2.137,P<0.05)。结论:高转移的结肠癌细胞在促使肿瘤血管生成能力上存在优势。

Comparative study in metastasis potential of human colon cancer cell lines

Objective:To clarify the main factors involved in the metastasis of colon cancer.Method:LS174 T and HCT 8,the two colon cancer cell lines different in potential of metastasis were separately detected in cell growth,invasion,expression of HLA,B-7,CD54 and CD24,potential to induce migration of vascular endothelial cell,and expression of vascular endothelial growth factor,by using in vitro models,flow cytometry,ELISA and western blot.Results:The difference between LS174 T and HCT 8 was focused on the ability in inducing the migration of vascular endothelial cells.The average of migration of endothelial cells induced by LS 174T was 154.6±37.9,and that induced by HCT 8 was 113.0±72.7(t=2.137,P<0.05).Conclusion:The dominant position in angiogenesis facilitates cancer metastasis of colon cancer cells such as LS174 T.
