嗅鞘细胞移植联合甲基维生素B12修复髓鞘组织的研究

摘    要:目的研究嗅鞘细胞(OECs)的培养及其对实验性变态反应性脑脊髓炎的治疗功用。方法用新生wistar大鼠嗅球培养OEC,移植组将OECs悬液注入实验性变态反应性脑脊髓炎大鼠的侧脑室;联合移植组在侧脑室注入OECs的基础上,腹腔注入甲基维生素B12;假手术组按以上方法注入等量生理盐水;观察两组大鼠运动功能评分、组织病理学变化。结果运动功能评分:联合移植组优于OECs移植组,以上两组均显著优于假手术组,差别有显著意义(P〈0.05);组织病理切观察:联合移植组髓鞘修复程度优于OECs移植组,以上两组髓鞘修复程度均显著优于假手术组。结论OECs移植可以促进自身免疫性脑脊髓炎的髓鞘修复,改善运动功能,OECs与甲基维生素B12对于髓鞘的修复有较好的协同作用。

Abstract:Objective] To develop a method for the culture of OECs and to evaluate the therapeutic effect of OECs transplantation on experimental allergic encephalomyelitis (EAE). Methods] OECs were cultured in the olfactory bulb of newborn wistar rat. OECs suspension was infused into the lateral cerebral ventriculus of AE rats, intraperitoneal methyl-vitamineB12 associated with above OECs suspension in combined transplantion group, and the saline with the same quantity was infused in pseudo-operation group. Afterwards, sport-function grade, histo pathological change were observed. Results] The sport-function grade was highest in combined transplantation, higher in OECs transplantation, and lowest in pseudo-operation(P <0.05). The best myeline repairment was seen in combined transplantation, OECs transplantation was better than pseudo-operation in histo-pathological slices (P <0.05). Conclusion] OECs transplantation may contribute to myeline repairment of EAE and improvement of sport function. OECs transplantation and methyl-vitamine B12 are synergic on myeline repairment.
