加味丹寿汤对双酚 A 染毒孕鼠妊娠丢失的实验研究

摘    要:目的:探讨补肾活血方———加味丹寿汤对双酚 A 染毒孕鼠模型妊娠丢失的影响。方法:10 周龄 SD 受孕大鼠随机分为补肾活血组、补肾组、模型组、阴性对照组与空白孕鼠组,每组各 10 只。自孕 0 d 起,补肾活血组、补肾组分别予加味丹寿汤、补肾方浓煎液灌胃,模型组、阴性对照组、空白孕鼠组均予生理盐水灌胃作对照。期间补肾活血组、补肾组、模型组于孕第 7 11天分别腹腔注射双酚 A 50 m g (kg· d) ^-1,阴性对照组于孕第 7 11 天腹腔注射等体积的芝麻油,各组均于孕第 18 天处死取材,计算胚胎吸收率、胚胎质量、活胎率。结果:与模型组相比,补肾活血组、补肾组均能明显降低胚胎吸收率,增加胚胎质量,提高雌激素受体及孕激素受体含量(P 〈 0.05),明显改善胎盘病理现象,但均以补肾活血组效果明显(P 〈 0.05)。结论:加味丹寿汤能提高雌激素及孕激素受体含量,改善胎盘病理现象,从而抵制双酚 A 染毒孕鼠胚胎吸收、增加胚胎质量,提高妊娠成功率。

Abstract:Objective :To explore the influence of Mordified Dan Shou Tang on pregnant rat models of fetal loss induced by bisphenol A (BPA). Methods:10-week-old SD pregnant rats were divided randomly into nourishing kidney and promoting blood flow group,nouris-hing kidney group, model group, negative control group and blank group. From the first day of pregnancy, rats in nourishing kidney and promoting blood flow group were given Mordified Dan Shou Tang through intragastric administration;rats in nourishing kidney group were given the formula of nourishing kidney;while rats in other groups were given the physiological brine respectively through intragas-tric administration. From the seventh to the eleventh day of pregnancy, nourishing kidney and promoting blood flow group,nourishing kidney group and model group were given multi-site intraperitoneal injection of BPA with a dosage of 50mg · (kg · d)^ -1 while negative control group were given multi-site intraperitoneal injection of sesame oil with the same volume. On the eighteenth day of pregnancy, rats in all the five groups were killed for samples to calculate fetal absorption rate ,fetal weight and living birth rate. Results:Compared with model group, nourishing kidney and promoting blood flow group and nourishing kidney group could dramatically reduce the fetal absorp-tion rate, increase fetal weight, enhance the content of estrogen receptor and progesterone receptor (P 〈 0.05 ) and significantly improve the placental pathological condition with nourishing kidney and promoting blood flow group displaying better effects in the above items ( P 〈 O. 05 ). Conclusion : Modified Dan Shou Decoction could dramatically increase the content of estrogen receptor and progesterone re-ceptor and improve the placental pathological condition, thus decreasing fetal absorbility of pregnant rats of BPA and increasing fetal weight and the chances of pregnancy.
