增殖细胞核抗原和P53在胎儿和成人皮肤中的表达特征及其意义

摘    要:目的 :探讨增殖细胞核抗原 (PCNA)和 P5 3蛋白在胎儿与成人皮肤组织中的表达特征及其可能的生物学意义。方法 :用免疫组织化学方法和病理技术确定这两种蛋白在不同胎龄的胎儿皮肤 (8例 )和成人皮肤(8例 )中的定位和表达量。结果 :在妊娠早期和中期胎儿的皮肤组织中 ,PCNA呈强阳性表达 ,而 P5 3蛋白表达很低。随着胎儿生长发育 ,PCNA蛋白的阳性细胞率逐渐降低 ,而 P5 3蛋白表达逐渐增强。在发育后期的胎儿和成人皮肤中 ,PCNA的阳性细胞率显著下降 ,阳性信号主要分布于角质形成细胞和毛囊上皮细胞的胞核内 ;P5 3呈阳性表达 ,蛋白颗粒定位于表皮基底层细胞和部分成纤维细胞中。结论 :在不同发育阶段皮肤组织中 ,PCNA和 P5 3蛋白可能通过影响细胞内 DNA合成和修复 ,调节皮肤组织的生长发育、结构和功能的维持以及损伤后的修复。

Abstract:Objective:To explore the localization and expression characteristics of proliferating cell nuclear antigen (PCNA) and P53 in fetal and postnatal skins underlining their biological significances.Methods:The expression intensity and distribution of PCNA and P53 were detected with immunohistochemistry and pathological methods in 8 cases of fetal skins with different gestational ages and 8 cases of postnatal skins that were obtained from patients in our hospital with agreement.Results:In skins from early and mid trimester of pregnancy,the expression intensity of PCNA was strong,while the positive signals of P53 were very weak.In skins from late trimester of pregnancy and from adult,the protein expression levels of PCNA was significantly decreased. Its positive signals were chiefly distributed in the nucleuses of keratinocytes and follicular epithelial cells.P53 content was significantly elevated.Its protein particles were mostly located in epidermal basal lamina cells and some fibroblast cells.Conclusions:The results indicate that PCNA and P53 are involved in the skin development in embryonic stage,the structural and functional maintenance in adult through influencing DNA synthesis and genome repair in cells.
