摘    要:目的 通过组织血浆酶原激活剂 (TPA)诱导K5 6 2细胞分化 ,探讨白血病的多药耐药性。方法 应用体外细胞培养技术 ,通过细胞形态、细胞化学染色观察细胞分化 ;用免疫组化 (IC)、流式细胞术 (FCM )、聚合酶链反应 (RT PCR)等技术检测P 糖蛋白 (P gP)的表达和功能及多药耐药基因 1信使核糖核酸 (mdr1mRNA)的表达。结果 TPA诱导K5 6 2细胞向单核 /巨噬细胞分化 ;在此分化过程中mdr1mRNA表达随作用时间延长而渐增加 ,P gP表达及功能增强。 结论 mdr1mRNA、P gP表达及功能在TPA诱导K5 6 2细胞向单核 /巨噬细胞分化过程中随细胞分化而上调

Abstract:Objective To observe the multidrug resistance gene 1(mdr1) mRNA expression during K562 cells differentiation induced by tissue plasminogen activator(TPA).Methods Incubation in vitro and cellular morphology were used to observe the differentiation characteristics of K562 cells, and flow cytometry and immunohistochemistry were used to test the Pglyloprotein(P-gP) function, and PT-PCR was used to assay the multidrug resistanle mRNA expression.Results The morphology of K562 cell tended to mature differentiation after the treatment with TPA, during which the rate of differentiation-induced K562 was 4.5 % and 95 % in control group and experiment group respectively; K562 cells with NSE staining showed positive, and inhibiting rate of NaF of K562 decreased from 86 % to 35.5 % in experiment group. The MDRr1 mRNA expression and the P-gP function both increased gradually during K562 cells differentiation induced by TPA.Conclusions TPA can induce K562 cells to differentiate into monocytes /macrophages.Mdbr1 mRNA expression and the P-gP function both increased gradually during K562 cells differentiation induced by TPA.
