流式细胞术检测复宫宁诱导异位子宫内膜细胞凋亡的研究

摘    要:目的 :从分子水平探讨中药制剂复宫宁治疗子宫内膜异位症的作用机理 ,观察其是否能诱导异位子宫内膜细胞发生凋亡。方法 :用手术移植法复制内异症的大白鼠模型 ,将其分成对照组、复宫宁组、丹那唑组及联合用药共 4组 ,采用灌胃给药 4周后 ,通过流式细胞仪观察异位子宫内膜细胞是否有凋亡出现。结果 :1 肉眼观察 ,用药组的移植物体积明显小于对照组 ,其异位病灶生长受抑制。 2 用药组异位子宫内膜细胞凋亡指数显著高于对照组 (P <0 0 1)。 3 在流式细胞仪直方图上 ,用药组均可见有凋亡峰 (即“亚G1”峰 ) ,且S期细胞明显增多 (P <0 0 1) ,提示细胞周期阻滞于S期而出现凋亡。对照组则没有明显凋亡峰出现。 4 复宫宁组 ,丹那唑组及联合用药组血清E2 水平均显著低于对照组 (P <0 0 5 )。结论 :本研究说明复宫宁能诱导异位子宫内膜细胞凋亡 ,其机制可能是通过降低血清E2 水平 ,致使细胞上bc1 2的表达下降。

Abstract:Objective:To explore the mechanism of chinese drugs Fugongning on cures of Endometriosis at the molecular level, and to investigate whether apoptosis is induced in endometriosis by the medicine. Methods: After making EMT animal model the rats were treated with Fugongning. Danazol and both medicines. Then apoptotic cells were detected by flow cytometry (FCM).Result:1. On gross examination the size of transplant in treated groups was significantly decresed as compared with that of control ( P <0 05). The growth of Endometriotic cells were inhibited. 2.The apoptosis index in Fugongning, Danazol and the both medicine group was significantly higher than that of control ( P <0 )1).3.After treatment with Fugongning, Danazol and both medicines, there were apoptotic peaks in cell cycle analysis on FCM. The cells in S phase were obvionsly increased ( P <0 01),which indicated that cell cycles were hindered in S phase. 4. The serum E 2 level in Fugongning, Danazol and the both medicine group was significantly lower that that of control ( P <0 05). Conclusion: This study indicates that Fugongning can induce apoptosis of Endometriotic cells. The possible mechanism is from decreasing the serum E 2 level to suppress bc1 2 expression in endometriotic cells.
