应用多细胞系微量细胞培养法分离脑炎病毒

摘    要:目的探索应用多细胞系微量细胞培养法进行病脑脑脊液病毒分离,以建立简便、快速的病毒初筛方法。方法采集病毒性脑炎病例脑脊液46份,应用4种细胞系在96孔细胞培养板进行分离病毒,所得培养物用肠道病毒通用引物RT-PCR扩增,进行初步鉴定。结果 46份标本分离病毒11株,其中3株应用肠道病毒通用引物扩增出预期片段。结论多细胞系微量细胞培养法具有快速、便捷的特点,是一种较好的病毒初筛方法。

Abstract:Objective Isolating virus from cerebrospinal fluids applying multi-cells and micro-plate method to build a simple and convenient method to initial screen virus.Method Collect 46 cerebrospinal fluids(CSF)from the viral encephalitis cases and isolate virus applying 4 cells in 96-well-microplate.The culture obtained were amplified with intestinal virus universal primer RT-PCR and identified preliminarily.Results 11 strains were isolated from 46 specimens,and 3 strains were amplified using consensus primers for enterovirus.Conclusions Multi-cells and micro-plate method was a convenient,simple and better method for initial screen virus.
