Interaction between digoxin and tricyclic antidepressants in the rat |

Abstract: | A possible interaction between digoxin and tricyclic antidepressants has been investigated in the rat. Acute administration of desmethylimipramine, 20 or 40 mg/kg, i.p., but not of imipramine, amitriptyline or nortriptyline, increased the lethality of i.v. infused digoxin in urethane anaesthetised rats. Chronic administration, 10 mg/kg/day for 14 days in the drinking water, of imipramine or desmethylimipramine, but not of amitriptyline or nortriptyline, also significantly increased digoxin toxicity. Mild stress did not significantly affect the toxicity of digoxin but stressing animals in the presence of amitriptyline or imipramine significantly increased the lethality of digoxin. Reserpine pretreatment afforded significant protection against digoxin. The role of catecholamines in this interaction is discussed. |
