人精子膜上凝集素受体的研究

摘    要:从糖专一性相同或不同的24种凝集素中筛选出SML、MDL、CKL和PHA-S四种能与人精子发生强烈反应的凝集素,浓度为0.98μg/ml时就能使精子凝集成块,6 h后,凝集团块也不分散,用机械方法分散团块所得精子已完全失去活动能力;其凝集作用可被凝集素各自的抑制糖所抑制;用FITC标记的SML、CKL、MDL与正常人精子作用后,在荧光显微镜下可观察到各凝集素受体的分布部位各不相同;用去垢剂增溶精子,Brij 58的增溶效果最好,增溶的膜蛋白可强烈地抑制凝集素对兔红细胞的凝集作用。

Abstract:Twenty-four lectins were tested for their ability to agglutinate human spermatozoa. Results showed that four lectins (SML. MDL. CKL and PHA-S) induced agglutination of human spermatozoa at a concentration of 0.98μg/ml. The agglutination of MDL, CKL and SML was inhibited respectively by N-acetyl-D-galactosamine, D-galactose and N-acetyl-D-glucosamine. We also found that four lectins (SML, MDL, CKL and PHA-S) at 0.9 mg or 1.2 mg could immediately agglutinate and kill all spermatozoa in 1ml semen, and the formed clots couldn't be liquefied after incubation for six hours at 36℃.Localization of MDL, SML and CKL receptors on the surface of human ejaculated spermatozoa was also investigated by FITC-lectins. The three lectins had different distributions on the sperm surface. SML receptors were mainly located on the equatorial region of spermatozoa heads. Location of CKL receptors was on the post nuclear region and neck. But MDL receptors were mainly on the post-acrosomal region. Few of them were found on the acrosomal cap and tail.The solubilization of human sperm membrane proteins treated with several detergents was examined. The result suggests that Brij-58 was the most efficient one of these detergents. Brij-58-solubilized sperm membrane proteins could strongly inhibit the agglutination of rabbit erythrocytes by lectins.
