早期自然流产蜕膜组织细胞凋亡与热休克蛋白70、P53的表达

摘    要:目的观察早期自然流产蜕膜组织细胞凋亡情况及与热休克蛋白70(HSP70)、P53表达的关系。方法采集20例正常早孕人工流产和20例早期自然流产的蜕膜组织,应用末端脱氧核糖核酸转移酶介导的缺口末端标记法(TUNEL)检测蜕膜组织细胞凋亡情况,免疫组织化学法检测蜕膜组织中热休克蛋白70、P53的表达。结果(1)早期自然流产组蜕膜与正常早孕蜕膜组织相比,凋亡显著增加(P〈0.01);(2)蜕膜组织中HSP70、P53表达,自然流产组与正常早孕组相比显著增强(P〈0.01)。结论蜕膜组织凋亡过度可能为自然流产发生机制之一,蜕膜细胞凋亡的增多与P53表达上调具有一致性;HSP70不能抑制蜕膜细胞凋亡,可能对蜕膜细胞有保护作用。

Abstract:Objective: To investigate the relationship between the apoptosis and expressions of heat shock protein70 (HSP70) and P53 in decidual tissue of early spontaneous abortion. Methods. The decidual tissue samples were obtained from 20 cases of surgical aspiration of early pregnancy and 20 cases of early spontaneous abortion. Apoptosis was detected with terminal deoxynucleotidyl transferas-mediated dUTP-biotin nick ending labeling(TUNEL). Expressions of Hsp70 and P53 proteins were detected with immunohistochemistry. Results: The apoptosis indices decreased in decidual tissue of spontaneous abortion than that of surgical aspiration (P 〈 0. 01). Hsp70 and P53 expressions were decreased in decidual tissue of spontaneous abortion than those of surgical aspiration (P〈0.01). Conclusions: Abnormal apoptosis in decidual tissue might be one of the mechanisms of early spontaneous abortion. Increased apoptosis in decidual tissue of early spontaneous abortion was coincident with the up-regulation of expression of Hsp70. The higher expression of Hsp70 could not suppress apoptosis in decidua.
