新加良附方联合5-Fu对裸鼠胃癌移植瘤Bax/Bcl-2表达的影响

摘    要:目的观察新加良附方联合5-Fu对人胃癌细胞(SGC-7901)裸鼠移植瘤生长抑制作用及对肿瘤组织Bcl-2及Bax蛋白表达的影响。方法建立移植性人胃癌细胞(SGC-7901)裸鼠移植瘤模型,随机分为模型对照组、5-FU组及新加良附方高、中、低剂量组、联合给药组。连续给药10d。计算肿瘤抑制率,检测Bax/Bcl-2蛋白表达。结果联合给药组抑瘤率为70.07%,与模型对照组比较,差异有统计学意义(P<0.05);与5-Fu组和新加良附高剂量比较,联合给药组抑瘤率明显升高,差异有统计学意义(P<0.05);联合给药组上调肿瘤组织Bax蛋白表达,下调Bcl-2蛋白表达,与模型对照组比较,差异有统计学意义(P<0.05,P<0.01);与5-Fu组和新加良附高剂量组比较,联合给药组Bax蛋白表达升高明显(P<0.05),Bcl-2蛋白表达下降明显(P<0.05)。结论联合给药组抑瘤率高于单独给药组,在下调Bcl-2、上调Bax表达方面优于单独给药组,显示两药协同作用的优势。

Abstract:Objective To observe the impacts of Xinjialiangfu Formula(Formula)on the inhibition of the growth of human gastric cancer( SGC -7901 )and Bcl- 2 and Bax protein expressions in tumor tissue of the nude mice of xenograft. Methods The xenograft models of nude mice were prepared with the trans planting SGC - 7901 and randomized into a model control group, a 5 - Fu group, a high - dose Formula group, a middle - dose Formula group, a low - dose Formula group and a combined medication group. Medica tion was given continuously for 10 days. The tumor inhibition rate was calculated. Bax/Bcl -2 protein expres sions were detected. Results The tumor inhibition rate was 70.07% in the combined medication group, presenting the significant difference as compared with the model control group(P 〈 0.05 ). The tumor inhibi- tion rate was increased remarkably in the combined medication group as compared with that in the 5 - Fu group and high -dose Formula group, indicating the statistically significance (P 〈 0.05 ). Bax protein expres sion in tumor tissue was up - regulated in the combined medication group and Bcl - 2 protein expression was down - regulated, presenting the statistical significance as compared with the model control group ( P 〈 O. 05 ) ,P 〈 O. 01 ). Bax protein expression was increased remarkably and Bcl -2 protein expression was reduced apparently in the combined medication group as compared with that in 5 - Fu group and the high - dose For mula group(P 〈0.05). Conehtsion The tumor inhibition rate in the combined medication group is higher than that in the single medication group. And the down - regulation of Bcl - 2 expression and the up - regula tion of Bax expression are superior to the single medication group. It indicates the advantages of the co - ac- tion of two medications.
