掺锶硫酸钙骨修复材料体外细胞相容性研究

摘    要:目的验证掺锶硫酸钙复合骨修复材料的细胞毒性和细胞相容性。方法用a-半水硫酸钙(a—CSH)和氯化锶(SrClz)采用混合共结晶法制备6组不同掺锶量的掺锶硫酸钙骨修复材料,用MTT法来检测细胞增殖情况,对各组材料进行细胞碱性磷酸酶活性测定。结果MTT实验显示,细胞在各组掺锶硫酸钙材料浸提液中均正常增殖,培养7d时掺锶材料各组OD值均高于a—CSH组(P〈0.01);碱性磷酸酶活性测定结果显示,成骨细胞碱性磷酸酶活性在掺锶材料浸提液中增加(P〈O.05),且当掺锶量为0.5%时最明显。共培养结果显示,细胞能在各组掺锶硫酸钙材料上紧密贴附生长,共培养4d后,掺锶0.3%和0.5%材料上的细胞多于a—CSH组,但差异无统计学意义(P-0.061,P-0.057)。结论掺锶硫酸钙骨修复材料无细胞毒性且有较好的细胞相容性,成骨样细胞能在其上正常地贴附生长。

The in Vitro Compatibility Study of Strontium-Calcium Sulfate Compounds as Bioactive Bone Grafted Substitute

Abstract:Objective To investigate a strontium-calcium sulfate compounds and test its cytotoxicity and biocompatibility in vitro. Methods Preparing the completely mixed powders of a-calcium sulfate hemihydrate (a- CSH) and SrClz to make strontium-calcium sulfate compounds materials with 6 different concentrations. TheMTT method was used to test the cell proliferation and the ALP activity of each group with different leaching liquors was examined. Results The MTT test demonstrated that the OD value of leaching liquor of strontium compound materials has some beneficial outcomes with cell multiplication of ROS17/2.8 celt line. On the 7th day of culture the OD value of all the strontium-calcium sulfate compounds group was higher than a-CSH group (P〈0. 01). For osteoblast-like cells, strontium incorporated materials can also increase their ALP activity(P〈0.05 ), especially when containing 0.5 % strontium. On the compound materials the cells can live and grow well. More cells contacted on the compound materials containing 0. 3% and 0. 5% strontium than it contacted on a-CSH, although there was no statistical difference (P = 0. 061, P = 0. 057 respectively). Conclusion The new materials, having good cell compatibility,do no harm osteoblast-like cells in vitro,which can have normal and regular growth on them.
