维甲酸对小鼠骨髓基质细胞粘附分子表达和粘附功能的影响

摘    要:探讨体外维甲酸 (RA)对小鼠骨髓基质细胞 (BMSC)粘附分子ICAM 1和VCAM 1的表达及人脐血单个核细胞 (UCBMNC)对BMSC粘附率的影响。用流式细胞术方法检测 0 .1 ,1 .0和 1 0 .0 μmol/LRA处理后BMSC粘附分子ICAM 1和VCAM 1的表达 ,以MTT方法检测 3种浓度RA处理后UCBMNC对BMSC粘附率的改变。结果显示 ,1 .0和 1 0 .0 μmol/LRA能增加BMSC上ICAM 1的表达及UCBMNC对BMSC的粘附率。RA对VCAM 1的表达无明显影响 ;RA上调ICAM 1的表达与增加UCBMNC对BMSC的粘附率呈正相关 (r =0 .7883 ,P <0 .0 5)。结论 :体外RA能上调小鼠BMSC上ICAM 1的表达和增加UCBMNC对BMSC的粘附率 ,两者呈正相关 ,为阐明BMSC粘附分子与造血干细胞归巢的相关关系及RA促进脐血造血干细胞归巢提供了体外实验依据

Abstract:To explore the effect of retinoic acid (RA) on the expression of intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) in murine bone marrow stromal cell (BMSC) and adhesive rate of human cord blood mononuclear cell (UCBMNC) to BMSC in vitro, the express ions of ICAM-1 and VCAM-1 of murine BMSC were detected by flow cytometry and the binding capacity of UCBMNC to BMSC was tested by MTT assay after co-culturing with 0.1, 1.0 and 10.0 micro mol/L RA, respectively. The results showed that 1.0 and 10.0 micro mol/L RA increased the expression of ICAM-1 and the adhesive rate of U CBMNC to BMSC, however, RA did not induced the increase of expression of VCAM-1. It was positive correlation between the increments of ICAM-1 expression and the adhesive rate (r = 0.7883, P < 0.05). It is concluded that RA up-regulated ICAM-1 expression of BMSC and increased the adhesion of UCBMNC to BMSC in vitro. These may clarify the correlation between adhesion molecules on BMSC and homing of hematopoietic stem cells, and provide the experimental basis for RA to promote the homing of umbilical cord blood stem cell.
