摘    要:目的用指纹图谱的方式研究宁夏苦豆子各同器官不同物候期化学成分的动态变化规律。方法采用HPLC/UV检测方法,色谱柱C18(4.6 mm×250 mm,5μm);流动相为0.05 mol·L^-1的磷酸二氢钾溶液(用三乙胺调节pH至7.0)-乙腈,梯度洗脱;进样量10μl,检测波长为205 nm,流速1.0 mL·min^-1。结果不同物候期的10批苦豆子叶、茎、根和籽分别得到16、10、13和7个共有峰,通过与对照品比较槐定碱、野靛碱、氧化槐果碱、苦参碱、氧化苦参碱在4个器官中均含有,槐果碱和莱曼碱存在于苦豆子叶和根中,槐胺碱仅存在于苦豆子叶中,籽中未检测到苦豆碱。结论通过指纹图谱的分析体现了不同物候期苦豆子叶、茎所含化学成分及各成分的含量随采收时间的不同变化较大,而苦豆子根、籽变化不明显。

Abstract:Objective To study the changes of chemical compositions in Sophora alopecuroides L. of different growth time of all organs in Ningxia by the method of fingerprint. Methods Waters X-Brige C18( 4. 6 mm × 250 mm,5 μm) column was used. The acetonitrile-0. 05 mol /L KH2PO4 solution( 2. 0 mol·L^-1triethylamine) with gradient elution was used as the mobile phase and 1. 0ml /min as the flow rate. The length of detection wave was 205 nm. Results By using similarity calculation,the 10 batches samples in leaves,stems,roots and seeds of Sophora alopecuroides L. from different growth times were analyzed,and the sixteen,ten,thirteen and seven characteristic peaks were indicated on HPLC fingerprints. And the cytisine,sophoridine,oxymatrine,oxysophocarpine,matrine existed in all organs,and the sophocarpine and lehmann presented in the leaves and roots,and the sophoramine existed only in the leaves,aloperine was not detected in the seeds. Conclusion The component and content of compounds have changed in leaves and stems of Sophora alopecuroides L. from different growth times,however the changes of roots and seeds of Sophora alopecuroides L. are not obvious.
