肝龙不同有效部位体外抗氧化活性的分析

摘    要:目的对抗乙肝病毒中药肝龙不同有效部位(肝龙总提物、肝龙活性成分Ⅰ、肝龙活性成分Ⅱ)进行了体外抗氧化活性的初步分析。方法采用体外化学模拟体系研究了"肝龙"3个有效部位清除DPPH自由基、羟基自由基和超氧阴离子自由基的能力,以及对3个有效部位的还原能力、抑制亚油酸过氧化作用进行了测定。结果肝龙总提物、肝龙活性成分Ⅰ、肝龙活性成分Ⅱ清除DPPH自由基能力的EC50值分别为0.309,0.088 8和0.386 3 mg.ml-1;清除羟基.OH自由基的EC50值分别为0.043 5,18.60和0.151 7 mg.ml-1;总还原力的EC50分别为1.754,0.489和2.139 mg.ml-1;在β-胡萝素/亚油酸体系中,3个有效部位均可起到抑制亚油酸被氧化的作用。结论抗乙肝病毒中药"肝龙"不同有效部位都能起到一定的抗氧化作用,各自表现出不同的抗氧化能力,其对自由基的清除机制可能不同。

Abstract:Objective To study the antioxidant activity of total extracts of Ganlong,active ingredient I and active ingredient II in Ganlong.Methods The antioxidative properties of these compounds were evaluated by total reducing power,superoxide radical scavenging assay,hydroxyl radical scavenging assay and DPPH radical scavenging assay.Results The total extracts of Ganlong,active ingredient I and active ingredient II in Ganlong had scavenging activities on DPPH(EC50 value was 0.309,0.0888 and 0.3863 mg/ml,respectively),OH.(EC50 value was 0.0435,18.60 and 0.1517 mg/ml,respectively) and high and dose-dependent reducing power(EC50 value was 1.754,0.489 and 2.139 mg/ml,respectively).In β-carotene-linoleate system,three kinds of active ingredients in Ganlong showed the capacity to inhibit β-carotene bleaching.Conclusion The active ingredients in Ganlong have antioxidative activity,which might display various mechanisms of action for radical scavenging down-regaluted.
