葛根素联合运动对去卵巢大鼠早期骨代谢及E_2水平的影响

摘    要:目的研究葛根素联合中等强度运动早期干预去卵巢大鼠(OCR),对其骨代谢及血清E2水平的影响。方法取12周龄的雌性SD大鼠48只,随机分为6组(A-F组),8只/组,其中B-F组去除双侧卵巢,A组仅单纯打开腹腔不切除卵巢。术后1周,A、B组不加干预;C组予以E2200μg/(kg.d),肌肉注射;D组予以葛根素50 mg/(kg.d),腹腔注射;E组以中等强度运动干预;F组以葛根素联合运动干预,方法分别同D、E组,全部常规饲养。干预12周后,麻醉,取血离心(腹主动脉穿刺取血),测定大鼠血清中钙、磷含量(电极法)、E2水平(放免法);右胫骨上端HE染色,观察骨组织结构。结果 B组血Ca浓度高于C、D、F组,差异显著(P〈0.05);B组ALP浓度低于C、F组,有统计意义(P〈0.05)。HE染色见B组大鼠骨细胞较少,排列紊乱,骨小梁稀疏,小梁间隙很大,小梁有断裂现象,F组小梁连续性好,结构紧密,脂肪空泡少。结论葛根素联合中等强度运动促进成骨,与单独使用大剂量E2作用相近。

Abstract:Objective To observe the impact on early bone metabolism and serum E2 expression of ovariectomized rats(OCR) using puerarin combined with moderate intensity exercise.Methods Forty-eight 12-week female SD rats were randomly divided into 6 groups(A-F,8/group).Under 0.3% pentobarbital sodium(30 mg/kg) anesthesia conditions,rats of group B to F were opened abdominal and removed bilateral ovary and rats of group A were pure opened abdominal without removing bilateral ovary.One week after operation,rats of group A and B were administered nothing;rats of group C were administered estrogen 200 μg/(kg·d),intramuscular injection for 12 weeks;rats of group D were administered puerarin 50 mg/(kg·d),intraperitoneal injection for 12 weeks;rats of group E were intervened with moderate intensity exercise;rats of group F were treated with puerarin combined with sport.Conventional breeding was used to all of the rats.After treatment for 12 weeks,serum of the rats was collected and spined,and the content of Ca and P were analyzed using electrode technique,the level of E2 was analyzed by using radioimmunity method.Bone struction of right upper tibia were examined by using HE staining.Results The concentration of Ca in group B was higher than group C,D and F which had significant difference(P0.05) and the concentration of ALP in group B was lower than group C and F(P0.05).The ultrastructure of rats tibia were observed through HE staining.The number of bone trabeculae and thickness were decreased significantly in group B,and the bone structure changed from strong density cellular structure to sparse loose irregular appearance beehive structure.While in group F,bone trabeculae had good continuity,compact structure and less fat empty bubble.Conclusion Puerarin combined with moderate intensity exercise can promote bone-formation and has similar role to single high-dose E2.
