精神分裂症患者血浆及单核细胞中microRNA表达水平差异的研究

摘    要:目的 探讨精神分裂症患者血浆和外周血单核细胞内microRNA(miRNA或miR)表达水平的差异.方法 对连续入组60例精神分裂症患者和72例正常对照,使用实时荧光定量PCR技术检测病例组和对照组血浆和外周血单核细胞内9种miRNA(miR-30e、miR-34a、miR-181b、miR-195、miR-346、miR-432、miR-7、miR-132、and miR-212)的相对表达水平.结果 病例组血浆miRNA-132、miRNA-195、miRNA-30e、miRNA-7(5.06± 1.16、2.99± 1.27、1.88±1.62、9.59±1.85)较对照组(5.91±0.93、4.45±1.06、3.56± 1.30、11.8±2.53)显著上调(P<0.05 ~ P<0.001).病例组外周血单核细胞miRNA-212、miRNA-34a、miRNA-30e(0.28±0.54、2.69± 1.10、-4.72± 1.01)较对照组(0.84±0.64、3.97±1.01、-3.55±1.26)同样显著上调(P< 0.05~ P<0.001).ROC曲线分析显示,miR-30e在血浆和单核外周血中均具有较高诊断价值.从血浆提取的microRNA-30e的AUC是0.767 (95%CI:0.608~0.926),敏感度90.90%,特异度60.00%;从外周血单核细胞提取的miRNA-30e的AUC是0.756(95% CI:0.584~0.929),敏感度81.80%,特异度68.00%.结论 精神分裂症患者外周血血浆及单核细胞中microRNA的表达水平不同,无明显的相关性.miR-30在血浆和单核外周血中具有较高的诊断价值.

Partial microRNA profiling differences between peripheral plasma and mononuclear leucocytes in schizophrenia patients

Abstract:Objective To explore the differences of microRNA expression as specific blood biomarker between peripheral plasma and mononuclear leucocytes in schizophrenia patients.Methods A total of 60 schizophrenia patients and 72 healthy controls were enrolled into our study,and nine most reported schizophrenia-associated microRNAs (miR-30e,miR-34a,miR-181 b,miR-195,miR-346,miR-432,miR-7,miR-132 and miR-212) expression levels were tested in peripheral plasma and mononuclear leucocytes using real-time PCR.Results MiRNA-132,miRNA-195,miRNA-30e and miRNA-7 (5.06± 1.16,2.99± 1.27,1.88± 1.62,9.59± 1.85) showed significantly higher expression in plasma samples(5.91±0.93,4.45±1.06,3.56± 1.30,11.8±2.53) (P<0.05,P<0.001) ; miRNA-212,miRNA-34a and miRNA-30e(0.28±0.54,2.69± 1.10,-4.72± 1.01) also showed significantly higher expression in mononuclear leucocytes samples (0.84±0.64,3.97± 1.01,-3.55 ± 1.26) (P<0.05,P<0.001).ROC curve analysis suggested that the AUC of microRNA-30e in plasma was 0.767 (95% CI:0.608~0.926) with sensitivity of 90.90% and specificity of 60.00%,and the AUC of microRNA-30e in mononuclear leucocytes was 0.756 (95% CI:0.584-0.929) with sensitivity of 81.80% and specificity of 68.00%.Conclusion The microRNA expression in plasma varies greatly with that in mononuclear leucocytes.MicroRNA-30e can serve as specific bloodbased biomarker for schizophrenia diagnosis.
