氯沙坦对自发性高血压大鼠肾脏组织Bcl-2和Bax表达的影响

摘    要:目的:探讨氯沙坦对自发性高血压(SHR)大鼠肾脏组织Bax、Bcl-2表达的影响。方法:将40只11周龄雄性SHR大鼠随机分成4组:氯沙坦组氯沙坦30 mg/(kg.d),灌胃]、培哚普利组培哚普利2 mg/(kg.d),灌胃]、氨氯地平组氨氯地平6 mg/(kg.d),灌胃]和对照组各10只,另设11周龄雄性Wistar-Kyoto(WKY)大鼠作正常对照组(n=10)。实验前及实验开始后测大鼠尾动脉收缩压1次/4周。饲养并灌胃8周后,收集肾脏组织,采用荧光定量PCR法和免疫组化法检测各组肾脏细胞凋亡因子Bax、Bcl-2表达的情况。结果:氯沙坦、培哚普利、氨氯地平均能显著降低SHR大鼠的收缩压,各组间差异无统计学意义。其中氯沙坦能显著降低SHR大鼠肾脏组织中Bax的表达(P〈0.01),增加Bcl-2的表达(P〈0.01),而培哚普利组、氨氯地平组无明显作用。结论:三种降压药均有明显的降压作用,其中氯沙坦在高血压病程中可抑制细胞凋亡因子Bax和促进Bcl-2的基因及蛋白质表达。

Abstract:Objective: To investigate the effects of losartan on the expression of apoptotic regulatory proteins,Bcl-2 and Bax in the kidneys of spontaneously hypertensive rats.Methods: Sex and age matched SHR were randomized to receive following approaches losartan treatment(30 mg/kg-1·d-1 by gavage),perindopril treatment(2 mg/kg-1·d-1 by gavage),amlodipine treatment(6 mg/kg-1·d-1 by gavage)and placebo,each group contained 10 rats.Wistar-Kyoto rats were(WKY)(n =10) as control groups.Systolic blood pressure was measured by a tail-cuff method before and every four weeks during experiment.Kidney was studied using realtime RT-PCR and immunochemistry methods for the assessment of the level of Bax and bcl-2 genes and proteins expression.Results: Losartan,perindopril,amlodipine could significantly reduce systolic blood pressure of SHR.In which,losartan can significantly reduce the expression of Bax(P <0.01)and increased Bcl-2 expression(P <0.01)of the SHR kidney tissues.The perindopril group and amlodipine group had no significant effect.Conclusion: Three antihypertensive drugs have significant antihypertensive effects.Losartan has the effects in apoptosis factor of the inhibition of Bax and promotion of Bcl-2 in the expression gene and protein expression.
