趋化素对大鼠血管平滑肌细胞生物学行为的实验研究

摘    要:目的探讨趋化素对大鼠胸主动脉平滑肌细胞生物学行为的作用。方法利用脂质体2000及PBS转染大鼠胸主动脉血管平滑肌细胞(VSMC),联合空白对照组A、阴性对照组B及抑制组C,观察其对细胞增殖、迁移和凋亡的影响。结果在增殖实验中,C组在24 h后,光密度值为0.62±0.11均低于A组(0.92±0.16)和B组(0.89±0.12),差异有统计学意义(P<0.05);在细胞迁移实验中,C组的细胞数(0.35±0.08)显著少于A组(3.11±0.61)和B组(2.97±0.83)(P<0.05);A、B、C组的平均细胞凋亡率分别为(5.5±0.7)%、(6.3±1.0)%和(14.2±3.7)%,C组的凋亡率分别高于A组和B组(P<0.05)。结论沉默趋化素能够显著抑制大鼠VSMC的增殖和迁移,并能提高其凋亡水平,为血管增生性疾病的防治提供实验依据。

Abstract:Objective To observe the effects of chemerin siRNA interference on the biological behavior of vascular smooth muscle cell (VSMC) of thoracic aorta.Methods Liposome2000 and PBS were applied to transfect rat aortic vascular smooth muscle cells (VSMC),the effects on proliferation,migration and apoptosis were analyzed by enzyme linked immunosorbent assay(ELISA) in comparison to blank control group A,negative control group B and inhibition group C.Results The OD value of group C after 24 h was 0.62 ± 0.11,lower than both group A (0.92 ± 0.16) and group B (0.89 ± 0.12),(P < 0.05) in proliferation test.The cells number of group C was significantly less than group A and group B (P <0.05) in migration test.The average cell apoptosis rate of the three groups was (5.5 ± 0.7) %,(6.3 ± 1.0) % and (14.2 ± 3.7) % respectively.The apoptosis rate of group C was higher than that of A and B group(P < 0.05).Conclusion Chemerin inhibitor could induce growth inhibitory,migration inhibitory and increased apoptosis in VSMC,which provides experimental references for the prevention and treatment of vascular proliferative diseases.
