摘    要:目的探讨苗勒氏管抑制因子(AMH)对人卵巢黄素化颗粒细胞分泌功能的影响。方法体外受精-胚胎移植(IVF—ET)患者经阴道超声引导下采卵,加入基因重组人苗勒氏管抑制因子(rhAMH)作用于颗粒细胞,测定培养液雌二醇(E2)、孕酮(P)水平。结果在无促卵泡生成激素(FSH)作用下,rhAMH对颗粒细胞分泌E2、P无明显影响(P〉0.05),加入FSH后rhAMH明显抑制颗粒细胞E2分泌(P〈0.01)。FSH对颗粒细胞分泌AMH无明显影响(P〉0.05)。结论卵巢颗粒细胞自身分泌AMH且不受FSH的影响,AMH抑制FSH促进卵巢颗粒细胞分泌雌激素的作用,减少卵巢始基卵泡的耗竭,进而增加卵巢的储备能力。

The effects of AMH on the function of human granulosa cells in vitro

Abstract:Objective To investigate the effects of anti - mullerian hormone ( AMH ) and on the E2 secretion of human granulosa cells in vitro. Methods Granulosa cells were obtained from infertile patients undergoing IVF - ET cycles and cultured in M 199 + 10% FBS medium. E2/P contents in the culture media were detected by ELISA kit. Results There was no great significance of rhAMH on the E2 secretion in the absence of rhFSH, while it obviously decreased the E2 secretion with the co - effect of rhFSH (P 〈0.01 ). There was no significant difference of AMH between in absence of rhFSH and presence of rhFSH groups ( P 〉 0.05 ). Conclusion AMH influences the steroidogenesis of granulosa cells in vitro, and may play an important role on the growth and development of follicles in the ovaries of women.
