二十二碳六烯酸对低氧性肺动脉高压大鼠肺动脉平滑肌细胞凋亡的影响

摘    要:目的:观察二十二碳六烯酸(DHA)在动物整体水平的抗大鼠肺动脉高压作用及其对肺动脉平滑肌细胞凋亡的影响。方法:将雄性SD大鼠随机分为常氧组、低氧组、低氧DHA处理组,每组7只。大鼠缺氧肺动脉高压模型采用慢性间歇性低氧方法建立,通过右心导管法测定大鼠平均肺动脉压,精确称左右心室质量计算右心室指数,测定肺动脉平滑肌细胞凋亡率和Caspase3、Caspase9活性。结果:与常氧组比较,低氧组大鼠平均肺动脉压和右心室指数均明显升高(PPPP结论:DHA具有抗大鼠肺动脉高压作用,其机制与促进肺动脉平滑肌细胞凋亡有关。

Effect of docosahexaenoic acid on pulmonary artery smooth muscle cells apoptosis in hypoxic pulmonary arterial hypertension rats

Abstract:Objective:To observe the role of docosahexaenoic acid( DHA) on anti-pulmonary hypertension in rats and on apoptosis in pulmonary artery smooth muscle cells. Methods:Male Sprague-Dawley(SD) rats(200 to 250g) were randomly divided into control normoxic group,chronic intermittent hypoxic group and DHA-treated group. The hypoxic pulmonary hypertension model was made by chronic intermittent hypoxic method,mean pulmonary arterial pressure was measured by right heart catheterization,the right ventricle index was calculated through weighing the left and right ventricles accurately,the apoptosis rate of pulmonary artery smooth muscle cells was determined by TdT-mediated dVTP nick-end labeling method,Caspase 3 and Caspase 9 activities were measured by activity assay kits. Results:Compared with the control normoxic group,in hypoxia group,mean pulmonary arterial pressure and right ventricular index were significantly increased(P<0. 01),the apoptosis rate of pulmonary artery smooth muscle cells,the activities of Caspase 3 and Caspase 9 were significantly decreased ( P <0. 01 );Compared with the hypoxia group, mean pulmonary arterial pressure and right ventricular index in hypoxia DHA treatment group were decreased(P<0. 01),apoptosis rate of pulmonary artery smooth muscle cells, Caspase 3 and Caspase 9 activities were increased(P<0. 01). Conclusions:DHA has the anti-pulmonary hypertension effect in rats, the mechanism may be associated with promoting apoptosis happening of pulmonary artery smooth muscle cells.
