摘    要:目的探讨阿柏西普对大鼠角膜碱烧伤后新生血管的抑制作用。方法制备SD大鼠角膜碱烧伤动物模型,随机分成4组,每组各9只,A组、B组与C组分别给予20 g·L-1、25 g·L-1、30 g·L-1的阿柏西普滴眼液,D组为生理盐水组(给予生理盐水),每组每天2次滴眼治疗,共14 d。分别于碱烧伤后第3天、7天、14天,观察角膜新生血管(corneal neovascularization,CNV)情况并计算CNV面积。碱烧伤后第14天,处死全部小鼠,进行组织学和免疫组织化学检测各组CD34、VEGF蛋白表达情况。结果碱烧伤后第3天、7天和14天,A组、B组与C组的CNV面积均小于D组,差异均有统计学意义(均为P<0.05);B组与C组在各时间点上CNV面积差异均无统计学意义(均为P>0.05);但B组和C组与A组相比,差异均有统计学意义(均为P<0.05)。HE染色及免疫组织化学染色结果显示:角膜碱烧伤后第14天,各组角膜CD34和VEGF蛋白表达增强,与A组、B组、C组比较,D组CNV旺盛致密,CD34和V...

Abstract:Objective To investigate the inhibitory effect of Aflibercept on neovascularization after corneal alkali burns in rats.Methods Animal models of corneal alkali burns in SD rats were prepared and randomly divided into 4 groups,9 rats in each group.Rats in group A,B and C respectively received 20 g·L^-1,25 g·L^-1 and 30 g·L^-1 Aflibercept eye drops,those in group D received normal saline eye drops,twice a day for 14 days.Corneal neovascularization(CNV)was observed and CNV area was calculated on the 3rd,7th and 14th day after alkali burn.On the 14th day after operation,all rats were sacrificed,and protein expression of CD34 and VEGF in each group was determined by histology and immunohistochemistry.Results On the 3rd,7th and 14th day after alkali burns,CNV areas in group A,B and C were smaller than that in group D,and the differences were statistically significant(all P<0.05).No statistical difference was found in CNV area between group B and group C at various time points(all P>0.05),but differences were found in CNV area for group B or C comparing with group A(all P<0.05).On the 14th day after alkali burns,HE staining and immunohistochemical staining showed that protein levels of CD34 and VEGF increased in cornea of rats in each group.Compared with group A,B and C,CNV was vigorous and dense,and protein levels of CD34 and VEGF were high in group D.Compared with group B and C,there were more CNV and higher protein levels of CD34 and VEGF in group A.There was no significant difference in the distribution of brown particles between group B and group C.On the 14th day after alkali burns,the results of enzyme-linked immunosorbent assay showed that expression levels of TNF-αprotein in aqueous humor of groups A,B and C were significantly lower than that in group D,and the differences were statistically significant(all P<0.05).Statistical difference was found in expression level of TNF-αprotein in aqueous humor for group B and C comparing with that in group A(both P<0.05),but there was no significant difference between groups B and C(P>0.05).Conclusion Aflibercept has an inhibitory effect on CNV in rats after alkali burn,and 25 g·L^-1 Aflibercept group has the best therapeutic effect.
