阿那白滞素在治疗大鼠前节内眼模拟手术诱发的血-视网膜屏障破坏中的作用

摘    要:目的 探讨阿那白滞素在治疗前节内眼模拟手术诱发的血-视网膜屏障(blood-retinal barrier,BRB)破坏中的疗效。方法 将大鼠随机分为对照组、模型组、局部治疗组和全身治疗组,每组36只。造模后4 h、1 d、2 d、3 d、5 d和7 d,采用ELISA法检测房水和玻璃体内白细胞介素-1β(interleukin-1β,IL-1β)的浓度。采用Evans蓝作为示踪检测BRB的完整性。结果 造模后1 d、2 d、3 d、5 d四组间房水和玻璃体内IL-1β浓度差异均有统计学意义(均P<0.001)。与对照组相比,模型组、局部治疗组和全身治疗组房水和玻璃体内IL-1β浓度在造模后4 h略有增加(均为P>0.05),造模后1 d显著增加(均为 P<0.05),造模后2 d达到峰值(均为P<0.05),造模后3 d、5 d均下降(均为P<0.05),造模后7 d接近对照组水平(均为P>0.05)。局部治疗组和全身治疗组房水和玻璃体内IL-1β浓度在造模后1 d、2 d、3 d、5 d均低于模型组(均为P<0.05)。局部治疗组房水和玻璃体内IL-1β浓度在造模后1 d、2 d均低于全身治疗组,但造模后3 d高于全身治疗组(均为P<0.05)。造模后1 d、2 d、3 d、5 d四组间视网膜Evans蓝渗漏量差异均具有统计学意义(均为P<0.001)。视网膜Evans蓝渗漏量在造模后1 d局部治疗组低于全身治疗组(均为P<0.05),但在造模后2 d和3 d局部治疗组均高于全身治疗组(均为P<0.05)。局部治疗组和全身治疗组视网膜Evans蓝渗漏量在造模后1 d、2 d、3 d、5 d均低于模型组,但均高于对照组(均为P<0.05)。结论 IL-1β在前节内眼模拟手术诱发的BRB破坏中起重要作用,阿那白滞素可以减轻BRB的破坏作用。

Abstract:Objective To explore the role of interleukin 1β(IL-1β)in blood retina barrier(BRB)breakdown induced by anterior intraocular simulation operation,and the efficacy of Anakinra in the treatment of BRB breakdown induced by anterior intraocular simulation operation.Methods The rats were randomly divided into control group,model group,local treatment group and systemic treatment group,36 rats in each group.ELISA method was used to measure concentrations of IL-1βin aqueous humor and vitreous body 4 h,1 d,2 d,3 d,5 d and 7 d after modeling.Evans blue test was adopted to detect BRB integrity.Results There were statistical differences in IL-1βat aqueous humor and vitreous body among four groups 1 d,2 d,3 d and 5 d after modeling(all P<0.001).Compared with control group,concentrations of IL-1βin aqueous humor and vitreous body slightly increased 4 h after modeling(all P>0.05),obviously increased 1 d after modeling(all P<0.05),peaked 2 d after modeling(all P<0.05),decreased 3 d and 5 d after modeling(all P<0.05),and were close to control levels 7 d after modeling(all P>0.05)for rats in model group,local treatment group and systemic treatment group.Concentrations of IL-1βin aqueous humor and vitreous body for rats in local treatment group and systemic treatment group were lower than those in model group 1 d,2 d,3 d and 5 d after modeling(all P<0.05).Compared with systemic treatment group,concentrations of IL-1βin aqueous humor and vitreous body for rats in local treatment group were lower 1 d and 2 d after modeling,and higher 3 d after modeling(all P<0.05).Statistical differences were found in retinal Evans blue leakage among four groups 1 d,2 d,3 d and 5 d after modeling,respectively(all P<0.001),but no difference was found 4 h and 7 d after modeling(all P>0.05).Compared with systemic treatment group,retinal Evans blue leakage in local treatment group was lower 1d after modeling,and higher 2d and 3d after modeling(all P<0.05).Retinal Evans blue leakage in local treatment group and systemic treatment group were lower than those in model group and higher than those in control group 1 d,2 d,3 d and 5 d after modeling(all P<0.05).Conclusion IL-1βplays an important role in BRB breakdown induced by anterior intraocular simulation operation,and Anakinra can alleviate BRB breakdown.
