P53和P63蛋白在人骨肉瘤组织及恶性转化细胞株中的表达变化及意义

摘    要:目的探讨人骨肉瘤组织及经硫酸镍恶性转化的永生化骨肉瘤样细胞系HOS中P53、P63蛋白的表达及意义.方法采用免疫组织化学S-P法和图像分析技术检测35例骨肉瘤标本、经硫酸镍恶性转化前后的永生化骨肉瘤样细胞系HOS和永生化的人胚成骨细胞系hFOB中P53、P63蛋白的表达.结果 P53、P63在35例骨肉瘤标本中的阳性表达率分别为42.9%、25.7%,不同组织学类型的骨肉瘤标本中阳性表达率无明显差异(P>0.05),P53蛋白和P63蛋白阳性患者与其阴性患者间总生存率比较在统计学上差异无显著性(P>0.05);在正常对照的人胚成骨细胞系hFOB中P53蛋白未见明显表达,P63蛋白表达呈弱阳性,未转化的HOS细胞株中可见P53、P63表达,而经硫酸镍恶性转化的HOS细胞株中P53、P63表达水平明显增高,差异有统计学意义(P<0.05).结论 P63蛋白及突变型P53蛋白的过表达在硫酸镍诱导的HOS细胞株恶性转化过程中起重要作用,是骨肉瘤发生早期的重要分子事件.

Abstract:Objective To study the significance of P53 and P63 expression in the tissues of human osteosarcoma and the immortal osteosarcoma-like cell line HOS malignantly transformed by niekel sulfate. Methods Immunohistochemistry S-P method and computerized image analysis were used to detect the expressions of P53 and P63 in 35 human osteosarcoma tissues, an immortal non-tumoricgenic osteosarcoma-like cell line HOS, the HOS transformed by niekel sulfate and a conditionally immortalized human fetal osteoblastic cell line hFOB. Results The positive rate of P53 and P63 was 42.9% and 25.7% respectively in 35 human osteosarcoma tissues. There was no significant difference between the protein expression and the pathological typing in osteosarcoma (P>0.05). There was no difference in overall survival rate between P53 or P63 positive patients and the negative counterparts (P>0.05). In hFOB, the expression of P53 was not detected, and that of P63 was weakly positive. The expression of P53 and P63 in HOS transformed by niekel sulfate were higher than that in normal HOS (P<0.05). Conclusion The overexpression of P63 and mutant P53 may play an important role in malignant transformation of osteosarcoma cell line induced by niekel sulfate, and may act as an early event in tumorigenesis of osteosarcoma.
